SUBJECT①Copy the disease model of hypertensive rat withLiver-Yang Hyperactivity Syndrome and the disease model ofIschemia Liver-Yang Forming Wind Syndrome and the disease model ofhemorrhagic Liver-Yang Forming Wind Syndrome.②To comparemRNA expression of Ubiquitin carboxy-terminal hydrolase L1(UCH-L1) mRNA and steroidogenic acute regulatory protein(STAR)mRNA in the normal contral group, hyperactivityof liver yang contralgroup,the Ischemia Liver-Yang Forming Wind contral groups andHemorrhagic Liver Forming Wind groups, and explore the relatetive anddifference when the disease developmenting from hyperactivity ofLiver-yang to Liver-Yang forming wind.METHODS Randomly selected 10 rats form 55 male SD rats asnormal contral group,and remnant 45 rats used by Liver-YangHyperactivity Syndrome model building made by fuzi decoction andhigh density Nacl for four weeks. Observering the degree of irritationand conjunctiva hyperemia and measuring the blood pressure androtation time. To eliminate rats unsuccessful model 10 rats. The remnant36 rats expert 10 for Liver-Yang Hyperactivity the other 26 wererandomly selected to shape ischemia liver forming wind with MCAO (middle cerebral artery occlusion) and hemorrhagic liver forming windwithⅦtype collagenase stereotaxically injected into the globas pallidusof rats. TTC and HE dyeing methord check the model. The model ofsuccess was put to death after 24h. RT-PCR was used to determine theexpression of UCH-L1 mRNA and StAR mRNA in the four group.RESULTS1. state of health: 36 of 45 rats copy the Liver-Yang hyperactivitySyndrome model successfully, successful rate is 80% .The Liver-Yanghyperactivity Syndrome model group with high aggression andconjunctiva hyperemia and high blood pressure and rotation timeshorten.①aggression degress:After four weeks, aggression amongLiver-Yang Hyperactivity group ,the Ischemiativer Liver-Yang FormingWind groups and Hemorrhagic Liver-Yang Forming Wind group is moreobviours than the normal contral group (P<0.01), compared to heLiver-Yang hyperactivity Syndrome model group, the IschemialiverLiver-Yang Forming Wind groups and Hemorrhagic Liver-YangForming Wind group is higher (P<0.05).②. The success rate of Ischemialiver Liver-Yang Forming Windmodel and Hemorrhagic Liver-Yang Forming Wind model is 77% ,①The score of nerve function defect: according to Zealong' methord: if thescore is two and above, the cerebral ischemia model and theintracerebral hemorrhage model is success.②. The rest of TTC and HE: TTC dyeing show ischemic region of hemisphere is pale and the otherpart is red in cerebral ischemia model. HE dyeing show a lot of red cellinfiltrate around haematoma, nerve cell obvioursly decrease and muchleukomonocyte cell increase in the intracerebral hemorrhage model.3. The rest of RT-PCR:①compared to the normal contral group,UCH-L1 mRNA expression is weaker in Liver-Yang Hyperactivitymodel group ,the Ischemialiver Liver-Yang Forming Wind groups andthe Hemorrhagic Liver-Yang Forming Wind group (P<0.05),comparedto Liver-Yang Hyperactivity model group, UCH-L1 mRNA expression isweaker in the Ischemialiver Liver-Yang Forming Wind groups andHemorrhagic Liver-Yang Forming Wind group (P<0.01).②comparedto the normal contral group, StAR mRNA expression is stonger inLiver-Yang Hyperactivity model group ,the Ischemialiver Liver-YangForming Wind groups and the Hemorrhagic Liver-Yang Forming Windgroup (P<0.05), compared to Liver-Yang Hyperactivity model group,StAR mRNA expression is weaker in the Ischemialiver Liver-YangForming Wind groups and the Hemorrhagic Liver-Yang Forming Windgroup (P<0.01).CONCLUSION1. Copy the Liver-Yang Hyperactivity model,IschemialiverLiver-Yang Forming Wind model and Hemorrhagic Liver-Yang FormingWind model successfully. 2. UCH-L1 mRNA,StAR mRNA decrease in the Liver-YangHyperactivity Syndrome group and Liver-Yang Forming WindSyndrome group, and the degress of express is significant (p<0.05 orp<0.01) show Liver-Yang Hyperactivity Syndrome and Liver-YangForming Wind Syndrome have the same material,but Liver-YangHyperactivity Syndrome and Liver-Yang Forming Wind Syndrome aredifferent Syndrome, the different degress of gene express show that theyare different Syndrome characterative. UCH-L1 mRNA,StAR mRNAin Ischemialiver Liver-Yang Forming Wind groups and HemorrhagicLiver-Yang Forming Wind group is insignificant (p>0.05),infer thatUCH-L1 up-regulation and StAR down regulation may be the distinctiveof er-Yang Forming Wind...
