Objective: To establish the chronic renal failure (CRF) model induced by Adenine inrats and explor the relationship between expression of a-SMA and renal interstitialfibrosis (RIF).Methods: Twenty rats were randomly divided into two groups; normal control andCRF model groups. The CRF model was induced by intragastric administration withAdenine(250mg/kg, once a day, from lth to 12th day; once two days, from 13th to24th day) in rats. The serum contents of BUN, Scr, Ca and P were determined. Thepathological changes and proliferative degree of fibrotic tissue in renal tissue wereobserved under light microscope after H-E and Sirus red staining. Expression ofa-SMA and typeⅣcollagen was determined by immunohistochemical SP methods.Results: Compared with the control group, the serum contents of BUN and Scr wassignificantly increased (p<0.05) and the Ca and P was dysequilibrium in the CRFmodel group. The fibrous tissue, positive cells of a-SMA and typeⅣcollagenmarkedly increased in renal interstitial in the CRF mode group.Conclusions: Adenine-induced rats CRF model was a appropriate model to study theCRF and RIF, and positive cells of a-SMA play a important role in development ofRIF.
