| Purpose:To research the effects of shenshuaifang(SSF) on adenine-induced renal failure inrats and elucidate its possible mechanisms.Material and method:1. Building methods: Made of adenine lavage methods model of chronic renal failuredecompensation period.2. Animal groups:Chronic renal failure rats model was successful,according to body masswere randomly divided into four groups, Renal failure model group, According to10ml/kglavage daily to give the same amount of saline solution; renal failure high dose group,24g/kgto fill the stomach twice a day for SSF; drug control group,to0.9g/kg to fill the stomachonce a day for Xiqingjiaonang; the normal group, o fill the stomach once a day for an equalamount physiological saline. The treatments of each groups last for13weeks.3. Model evaluation: Through the renal tissue in rat models of HE staining, to observe thechanges of renal tissue pathological morphology, to observe the evaluation model of thepreparation to success.4. SSF intervenes adenine intragastric administration due to the impact of the the whole stateof the model rats of CRF.Observing the activity status, body hair, activities, behavior, bodyweight changes of each group.5. SSF intervenes adenine intragastric administration due to the impact of the the bloodbiochemistry of the model rats of CRF.Detecting the heme, serum calcium, phosphorus,creatinine and blood urea nitrogen of each group.6. SSF intervenes adenine intragastric administration due to the impact of the TGF-beta1,MCP-1and mRNA expression of the model rats of CRF.By immunohistochemical staining method,in combination with the quantitative imageanalysis technology, we study the TGF-beta1protein expression changes in chronic renalfailure rats,and renal failure of the intervention of chronic renal failure induced by adeninelavage model rat TGF-beta1, MCP1expression mechanism.Through the method of RT-PCR, to combine with the quantitative image analysistechnology, study the TGF-beta1protein expression changes in chronic renal failure rats, and SSF intervenes adenine intragastric administration due to the impact of the TGF-beta1,MCP-1and mRNA expression of the model rats of CRF.Results:Serum urea nitrogen and serum creatinine values of CRF rats were higher significantly thanthose of normal rats respectively, and these values of SSF group were evidently lower thanthat in CRF group. Hemoglobin values of SSF high dose group were significantly increased tothe level of control group. Compared with model, the serum calcium levels of SSF group ratswere obviously increased, and the serum phosphorus reduced. Furthermore, the renalhistopathological changed were lessened in SSF-treated rats. The expressions of MCP-1andTGF-β1in renal tissue were significantly lower in SSF group compared with the CRF group,respectively.Conclusion:SSF may ameliorate the renal fibrosis in adenine-induced chronic renal failure in rats. Theeffct is probably related to down-regulated expressions of TGF-β1and MCP-1. The exactmechanism still needs to be studied further. |
