Objective:To investigate the effects of curcumin on bFGF induced proliferation of hRPE (human retinal pigment epithelium, hRPE) and the effects of curcumin and bFGF on the expression of B-cell lymphoma/leukemia-2 (Bcl-2) mRNA of hRPE.Methods: hRPE culture in vivo was divided into blank control group,control group and experimental group. The blank control group was add DMEM containing 1% fatal bovine serum. The control group was add DMEM containing 1% fatal bovine serum and 5ng/ml bFGF. The experimental group was add DMEM containing 1% fatal bovine serum,5ng/ml bFGF and curcumin at varying concentrations of 10mg/L,20mg/L,40mg/L into the cultured hRPE. All continued to culture for 24h and 48h.MTT assay were used to detect inhibiting rates of cultured hRPE. The expression of Bcl-2 mRNA of different group culture for 48h was studied by Reverse Transcriptase-Polymerase chain reaction(RT-PCR).Result:5ng/ml bFGF can significantly promote hRPE Proliferation than the blank control group(P<0.01).Three concentrations of curcumin can inhibitant hRPE proliferation at 24h(p<0.05). 20mg/L,40mg/L curcumin can inhibitant hRPE proliferation at 48h(p<0.05).When hRPE culture in vivo for 48h,blank control group express bcl-2 mRNA and 5ng/ml bFGF can significantly improve the expression of bcl-2 mRNA. The expression of bcl-2 mRNA of hRPE was decreased to some extent when treat with three concentrations of curcumin. Curcumin at concentrations of 20mg/L,40mg/L can significantly reduce the expression of bcl-2 mRNA than control group.(P<0.05)Conclusion: Curcumin can inhibitant bFGF Induced Proliferation of human RPE. Curcumin and bFGF can impact the expression of Bcl-2 mRNA of hRPE. The expression of bcl-2 mRNA of hRPE regulate by curcumin may play key role on curcumin inhibitant bFGF induced proliferation of human RPE.
