Object: To study NO level in peritoneal fluid of patients with endometriosis and effects on human sperm motility.To find the possible relationship between NO level and low pregnancy rate of endometriosis.Methods: Peritoneal fluid was aspirated from the cul-de-sac,uterovesical pouch. Endometriosis patients without infertility history were refered Group 1(EMS Group); endometriosis patients with infertility history including primary and second infertility were refered Group 2(EMS-associated infertility Group);patients with nomal pelvic were refered Group 3(control Group).Peritoneal fluid samples were collected from nineteen women of Group 1,sixteen women of Group 2,twenty-one women of Group 3.Then peritoneal fluid macrophages were segregated,cultivated, concentration of NO metabolin(nitrate and nitrite) were detected. At last sperm motility were detected after incubation together with culture fluid.Results : NO level of peritoneal fluid in EMS group and EMS-associated infertility Group were significantly higher than that of control Group (P<0.05), There was no significant in peritoneal fluid NO level of different stage in EMS(P>0.05).Meanwhile,after four hour incubation with primitive supernatant,the sperm motility rate decreased significantly in EMS group and EMS-associated infertility group than control group(p<0.05),and there was no significant between Group 1 and Group 2 of sperm motility(p>0.05).,after four hour incubation with culture fluid, the sperm motility rate decreased significantly in EMS-associated infertility group than control group and EMS group(p<0.05),and there was no significant between control group and EMS group of sperm motility(p>0.05).Conclusion :It is likely that elevation of peritoneal fluid NO level in patients with endometriosis may play a role in the pathogeneses of EMS-associated infertility...
