Background Former researchs indicated that Sedum sarmentosum Bunge could refrain the immunologic response mediated by T cells.Inflammatory bowel disease(IBD) had the over-accentuation of mmune response mediated by T cells and unspontaneous cure of inflammatory reaction.FOXP3 was a kind of transcription factor who could influence the development of regulatory T Cells(Treg)and refrain the immunologic function..Objective (1)To Study the expression of FOXP3,TGF-β1 and IL-2,IL-10 , damage activity index(DAI), common morphous damage index (CMDI), tissue damage index(TDI) on trinitrobenzene sulphonic acid (TNBS)-induced colitis and experimental colitis rats administered with Sedum sarmentosum Bunge. (2) To Study the correlation of above-mentioned data and pathogenesis of TNBS-induced colitis. (2)To investigate the effect and mechanism of Sedum sarmentosum Bunge on experimental colitis.Methods The S-D rats were divided into 3 groups (n=10):Normal control , experimental colitis and experimental colitis administered with Sedum sarmentosum Bunge(SsB). experimental colitis were induced by TNBS. The expression of TGF-β1 and FOXP3 in colonic mucosa were determined by immunohistochemistry and cytokine IL-2,IL-10 in peripheral blood serum were analyzed by enzyme-linked immunosorbent analysis.Results (1) The damage activity index(DAI),the common morphous damage index(CMDI) and the tissue damage index(TDI) of the experimental colitis rats administered with SsB were better than the experimental colitis rats(P﹤0.05).(2) There was decreased IL-10 in peripheral blood serum and low-expression of TGF-β1 ,FOXP3 in colonic mucosa, increased IL-2 in peripheral blood in experimental colitis,while increased TGF-β1,FOXP3 and IL-10,decreased IL-2 in experimental colitis administered with SSB(all P﹤0.05).(3)In experimental colitis FOXP3 correlated to IL-2 negatively, r=-0.922,P<0.001; FOXP3 correlated to TGF-β1 positively, r=0.881,P=0.001; FOXP3 correlated to IL-10 positively, r=0.942,P<0.001; FOXP3 correlated to CDMI negatively, r=-0.920,P<0.001;FOXP3 correlated to TDI negatively,r=-0.875,P<0.001.Conclusion (1)There were increased IL-2,DAI,CMDI,TDI on TNBS-induced colitis and decreased after administering with Sedum sarmentosum Bunge ,while decreased FOXP3,TGF-β1,IL-10 and increased after administering with Sedum sarmentosum Bunge.(2) FOXP3 correlated to TGF-β1,IL-10, IL-2, CMDI,TDI and TNBS-induced colitis maybe correlated to them. (3)Sedum sarmentosum Bunge maybe relieve the symptoms of experimental colitis through the changes of FOXP3,TGF-β1,IL-2 and IL-10.
