Objective:To investigate the protective effect of Sedum sarmentosum Bunge. against Lipopolysaccharide (LPS) and D-galactosamine (D-GalN) induced septic shock in mice and its chemical constituents.Method:①Male ICR mice were randomized into normal group, model group, dexamethasone group and four disparate of Sedum sarmentosum Bunge. treated with different doses of medicines. Randomized seven groups and each group has five mice. The model was made by intraperitoneal injected with D-GalN 700mg/kg and LPS 50μg/kg after one hour injected the medicine. First through observed the mice survival rate for 48 hours.②After selected, using the same method, Separated the aqueous extract of Sedum sarmentosum Bunge. and screening the effective sites for septicemia induced by LPS and D-GalN. Comprehensive use of silica gel column chromato graph,1.5 hours after D-GalN/LPS treatment, blood was collected from retro orbital venous plexus to measuring TNF-a and IL-2.eight hours after D-GalN/LPS treatment the blood was collected to measuring serum ALT, AST and NO. Selected the best site that could improve mice survival rate significantly and for the next specific mechanism research.③sing sephadex column chromato graphy; high performance liquid chromato graph (HPLC) and high-speed countercurrent chromato graphy (HSCCC) and many kinds of methods to separated n-butanol effective sites in aqueous extract of Sedum sarmentosum Bunge.④Macrophages in vitro, compounds at 50μM or 25μM dose, with LPS 100 ng/ml induced macrophages secrete NO, PGE2 and TNF-a, supernatants collected in each group were measured NO, PGE2 and TNF-a levels.Results:①In the experiment of mice survival rate,compared with model group, n-bu tanol sites in aqueous extract of Sedum sarmentosum Bunge. group 12 hours after was increased mice survival rate to 80%,24、36、48 hours after was increased mice survival rate to 60%. So insure the protective effect of Sedum sarmentosum Bunge. was n-butanol group.②The n-butanol sites reduced the levels of ALT and AST with statistical significance compared with model group. According to the vitro data determined n-butanol layer was the effective sites in aqueous extract of Sedum sarmentosum Bunge.③Using 1H-NMR、13C-NMR and other spectral techniques to identified seven compound of the effective structure from n-butanol effective sites.④The six monomer compounds have obvious inhibition to various kinds of inflammatory factors that bring about by LPS-induced macrophages RAW264.7. Thereby, to suppress RAW264.7 cell’s proliferation and secretion function,to achieve anti-inflammatory and protective sepsis effect.
