Objective:To observe the expression of PTEN and CyclinD1 in normal ovarian tissues, benign ovarian neoplasm,epithelial ovarian cancer and study their association with clinical pathological features in epithelial ovarian cancer.The aim was to investigate their relationship with the development,metastasis of epithelial ovarian cancer and the clinical significance of PTEN,CyclinD1 for the diagnosis and cure of epithelial ovarian cancer.Methods:The expression of PTEN and CyclinD1 protein was determined in 75 cases by immunohistochemical methods,including 20 cases of normal ovarian tissues,15 cases of benign ovarian neoplasm,40 cases of epithelial ovarian cancer.The level of PTEN and CyclinD1 expression in 40 epithelial ovarian cancer was analyzed against clinical pathological factors such as age,clinical stage,histological grade.The association of PTEN and CyclinD1 was studied,too.Results:1.The percentage of PTEN positively expression decreased from normal ovarian tissues(85%)to benign ovarian neoplasms(73.3%)to epithelial ovarian cancer (32.5%),(p<0.05).Among them,there were statistically significant between epithelial ovarian cancer and narmal ovarian tissues(p<0.05)and between epithelial ovarian cancer and benign ovarian neoplasms(p<0.05).But no significantly difference was found between narmal ovarian tissues and benign ovarian neoplasms (p>0.05).2.The positive expression of PTEN was significantly associated with differentiation and clinical stage,but was irrelevant to age of the patients,histological types and lymph node metastasis.PTEN was expressed significantly higher in high differentiation than low differentiation(p<0.05).The positive rate of PTEN in stageⅢ-Ⅳgroup was lower than that of stageⅠ~Ⅱgroup(p<0.05). 3.The expression of CyclinD1 was 5%,13.3%,77.5%respectively in normal ovarian tissues,benign ovarian neoplasms,epithelial ovarian cancer(p<0.05).The positive expression was statistically difference between epithelial ovarian cancer and narmal ovarian tissues(p<0.05)and between epithelial ovarian cancer and benign ovarian neoplasms(p<0.05).However,there were no statistical significance was found between narmal ovarian tissues and benign ovarian neoplasms(p>0.05).4.Statistical significance was foung between CyclinD1 and the differentiation, clinical stage and lymph node metastasis.CyclinD1 was expressed significantly lower in high differentiation than low differentiation(p<0.05).The positive rate of CyclinD1 in stageⅢ-Ⅳgroup was higher than that of stageⅠ-Ⅱgroup(p<0.05). The expression of Cyclin D1 protein in epithelial ovarian cancer was related to metastasis(p<0.05).There were no significant difference between CyclinD1 and the patients,histological types.5.The expression of PTEN was negative correlated with the expression of CyclinD1 in epithelial ovarian cancer(r_s=-0.521,p<0.01).Conclusion:1.The expression of PTEN was descended with the transformation from normal ovarian tissues,benign ovarian neoplasms,epithelial ovarian cancer.The expression level of PTEN was associated with differentiation and clinical stage.2.The expression of CyclinD1 was descended with the transformation from normal ovarian tissues,benign ovarian neoplasms,epithelial ovarian cancer.The expression level of CyclinD1 was associated with differentiation,clinical stage and lymph node metastasis.3.The expression of PTEN was negative correlated with the expression of CyclinD1 in epithelial ovarian cancer,which suggested that the abnormal expression of PTEN and CyclinD1 may play important roles in the carcinogenesis.
