ObjectiveTo explore the expression and clinical significance of P-glycoprotein(P-gp), Glutathione-transferase pi(GST-Ï€) and Topoisomeraseâ…¡(Topoâ…¡) in cervical cancer.MethodsUsing immunohistochemical methods to detect the expression of P-gp,GST-Ï€,Topoâ…¡in 20 normal cervix,30 cervical intraepithelial neoplasia(CIN),60 untreated cervical cancer ,as well as 10 recurrent cervical cancer, investigating their relationship with clinicopathologic characteristics and survival rate in cervical cancer.Resultsâ‘ In normal cervix, CIN, untreated cervical cancer and recurrent cervical cancer, the expression rate of P-gp was 25%,63%,85% and 100% respectively, the expression rate of GST-Ï€was 20%,70%,93% and 100% respectively, the expression rate of Topoâ…¡was 10%,60%,73% and 90% respectively. Positive expression of P-gp, GST-Ï€and Topoâ…¡was significantly higher in cervical cancer and CIN than that in normal cervix(P<0.05).â‘¡P -gp ,GST-Ï€, and Topoâ…¡co-expressed in cervical cancer, positive correlation between the expression of P-gp and GST-Ï€was observed(P<0.05), the same result was found between the expression of P-gp and Topoâ…¡(P<0.05),but there was no correlation between the expression of GST-Ï€and Topoâ…¡(P>0.05).â‘¢I n untreated cervical cancer, the expression of P-gp,GST-Ï€was not associated with FIGO stage, histological type and tumor grade(P>0.05),the expression of Topoâ…¡in adenocarcinoma was significantly lower than that in squamous carcinoma(P<0.05),but was not associated with FIGO stage and tumor grade(P>0.05).â‘£P atients with high expression of P-gp ,GST-Ï€had a lower rate of 3-year survival than that with low expression of P-gp ,GST-Ï€(P<0.05), no difference was found between low and high expression of Topoâ…¡(P>0.05). Univariate analysis revealed that FIGO stage, histological type, P-gp and GST-Ï€were closely related with prognosis in cervical cancer, multivariate analysis revealed that FIGO stage and histological type were independent factors for prognosis in cervical cancer. Conclusionsâ‘ P-gp, GST-Ï€and Topoâ…¡probably have a relation with occurrence and advance in cervical cancer.â‘¡Patients with high expression of P-gp, GST-Ï€had a poorer prognosis than that with low expression of P-gp, GST-Ï€, these results suggest that P-gp, GST-Ï€may be useful markers of poor prognosis in cervical cancer.â‘¢P-gp,GST-Ï€, and Topoâ…¡co-expressed in cervical cancer, thus, measurement of them simultaneous in cervical cancer could be useful in the design of chemotherapeutic protocols.
