Atherosclerosis(AS),nonalcoholic fatty liver dieases(NAFLD) and cardiovascular diseases are the most harmful to public health.The main pathogenic factors,especially the high level of blood cholesterol,is the disorder of lipid metabolism.How to regulate the blood lipid effectively is a hotspot on nutriology research.The clinic use of Opuntia dillenii Haw.is extensive.OPS is one of effective components in Opuntia dillenii Haw.that has medicative effect.This is to investigate the regulation of Opuntia dillenii Haw.polysaccharides on lipid and its mechanism in hyperlipidemic rats.Methods:SD hyperlipidemic rats were induced by feeding high lipid emulsion.OPS was given to hyperlipidemic rats through gavage for four weeks. During the course,experimental rats were investigated by observing the changes in body weight,water intake,food intake.And the fast blood totalcholesterol(TC), triglyceride(TG),were tested at the first week,the second week.And at the forth week,the blood lipid(TC,TG,HDL-C,LDL-C),the activity of LCAT and SOD, the content of the MDA and NO were also determined.Meanwhile,the content of lipid and MDA,the activity of HMG-CoA reductase in liver were studied. Furthermore,we made observation with HEstaining on effect of this formula on pathomor- phology of liver tissue.By the end of the test,pathomorphlogocal examination were carried out and VCAM-1 were determined by immunohistochemical in vein.The main results were as follows:(1) At the end of the second week,the level of TC in the OPSⅢgroup were significantly lower than the model group(P<0.01).Ultimate experiment,the level of TC,TG and LDL-C in the OPSⅢgroup were markedly reduced 78.7%,26.3%, 35.2%at the same time(P<0.01).Contemporarily,the activity of LCAT and osmotic fragility cures were improved by OPS.(2) Compared with model group,OPS may have effect on reducing lipidic aggradation in liver and activation of TC and TG,at same times decreasing activity of HMG-CoA reductase,promoting SOD activation of hepatocellular microsome and decreasing MDA content(P<0.01).Meanwhile the results of pathology of liver showed that OPS may have better effect on anti-fatty liver and treatment on lipoid hepatitis,and have been obviously superior to zhibituo in anti-fatty liver.(3) Compared with model group,the content of MDA were markedly reduced 30.9,and the activity of SOD,the content of NO were significantly increased 56.7%,75.2%in the OPSⅢgroup.There were evident difference between OPS group and model group in activity of SOD,content of MDA and NO(P<0.01).The mechanism of OPS on AS may related to protecting endothelial cell and inhibiting the inflammation of endothelial cell.(4) By observation on pathology of blood vessel andimmunohisto- chemistry of VCAM-1,this result speculated that MDA and ox-LDL-C considered as a domnous factor,inducing the damnification of endothelial cell,then activation and excretion a mount of cytokine and adherence factor.The latter promote leucocyteendothelial cell adhension and hyperplasia of smooth muscle.Consequently chronic inflammation of AS comes into being.The adjustment and improvement of the OPS on lipid metabolic disorder and membrane lipid peroxidative damage of hyperlipemia rats were significant.At the same time,OPS could treat or prevent disease efficiently such as hyperlipoidemia, AS,fatty liver and so on.It was proposed that OPS were worth further researching and clinical application.
