Objective Through the research on the dendritic cell(DC) subsets express rate in patients with chronic hepatitis C virus(HCV) infection and the healthy control,the effect on the express rate of DC subsets in the chronic HCV(CHC) patients with Interferon(IFN) treatment,to investigate the effect of DC subsets in HCV persistents chronically infected,to investigate the effect of DC subsets with IFN.Methods There are 50 objections in this research.30 patients with HCV chronic infection were selected and 20 healthy persons observed as control.The express rate of dendritic cell subsets was examined in whole blood by flow cytometry(FCM).The serum samples from patients were collected for the measurement of HCV-RNA by real time fluorescent quantitative polymerase chain reaction(RT-PCR),the HCV genotype detected by sequence.Compare the express rate of DC subsets in HCV patients with that in healthy control.Patients were treated with IFN.The express rate of dendrtic cell subsets were investigated before the treatment and after 4 weeks,12 weeks treatment, and between the patients groups with different ALT level,with different HCV-RNA content,as well as with different HCV genotypes related DC subsets express rate were studied.To investigate the change and effect of the express rate of DC subset in the process with the IFN treatment.Results①the express rate of DC subsets(DC1,DC2) in the group of patients was significant lower than that in the control group(P≤0.05 ).The patients with HCV-RNA≥2.0+E006 contents was significant lower than that with HCV-RNA<2.0+E006 contents. However,the difference of the express rate of DC subsets among the patients groups with different ALT level has no significant deviation(P>0.05).It had not done relevant statistical in the patients with different genotype②Patients who had received IFN treatment for 4 weeks,the express rate of DC1,DC2 count was significant lower than those of patients before treatment(P≤0.05);the difference of the express rate of DC subsets between the patients group after IFN treatment for 12 weeks and the same patients group before treatment was no statistical significant in DC1 count(P>0.05).The DC1,DC2 express rate in the patients whose HCV-RNA contents was not detected after the therapy with interferon for 4 weeks,12weeks was significant higher than those in patients whose HCV-RNA contents was detected after the therapy. Among those patients the group with different ALT level,has no significant deviation.The patients with different genotype and different HCV-RNA content level had not done statisticas.Conclusion There was immune dysfunctional in patients with chronic HCV infection.,the express rate of DC1,DC2 in the HCV patients was significant lower than that of healthy control.The express rate of DC subsets was influenced by HCV-RNA content in HCV patients.The DC1,DC2 express rate was effected by the therapy with IFN in HCV patients,as well as the function of DC subsets was better after the treatment by IFN.And its curative effect has no difference in the patients with different ALT level.
