Experimental Study On The Treatment Of Vascular Dementia Of Compound Gingko Leaf Tablet

Objective:To study the effects of Compound Gingko Leaf Tablet on Vascular Dementia animal models and investigate the mechanism of action.Methods:Established Vascular Dementia animal models and studied the ethology, the content of MDA in serum,the activity of SOD and AChE in brain tissues,and observed hippocampus pathomorphology of the animals.Established blood stasis models and insufficiency of kidney-YAND animal models based on traditional Chinese medicine differentiation of symptoms and signs and studied hemorheology, auricle minicirculation,the level of cylic nucleotides in blood plasma of the animals,etc.Results:The experiment results showed that the learning and memory ablilties of mice in Compound Gingko Leaf Tablet group were better than that of Model group. Compared with Model group the content of MDA in serum of animals in Compound Gingko Leaf Tablet group decreased,the activity of SOD enhanced, and the activity of AChE reduced in brain tissues.The result of hippocampus HE dyeing showed that nerve cells of hippocampus CA1 district in Compound Gingko Leaf Tablet group were intact,lining up in order,no necrosis obviously,while there are evidently pycnosis in Model group.Immunohistochemistry staining results displayed that the expression of BAX in hippocampus CA1 district in Compound Gingko Leaf Tablet group was weaker than that of Model group,but the expression of hippocampus SS was stronger than that of model group(P＜0.05). Compared with Model group the Blood viscosity,plasma viscosity and volume of packed red cells of rats in Compound Gingko Leaf Tablet group were reduced, arteriole and veinule of pinna aurium of mice were dilated obviously.The Yang asthenia symptom of rats and mice in Compound Gingko Leaf Tablet group were relieved significantly,and the level of cylic nucleotides in blood plasma have been adjusted,the difference was evident between Compound Gingko Leaf Tablet group and Model group.Conclusions:Compound Gingko Leaf Tablet can improve the learning and memory abilities of Vascular Dementia animal models.The experiment results may hint that this tablet may clean up free radical,reduce lipid peroxidation formation, increase neurotransmitter indirectly,protect hippocampus nerve cell,restrain brain post traum apoptosis,increase hippocampus SS expression,etc.And this tablet can promote blood flow,dissolve stagnant blood,enhance kidney-YANG,accordingly make the brain healthy and enhance wisdom.
