A Combination Of Different Prescriptions. Wu Tang Role Of Vascular Dementia In Rats

Object:Based on VaD can be ameliorated by Siwutang,the purpose of this research is to explore the key constitutes of Siwutang(Decoction of four herbs, Angelica sinensis(Oliv.)Diels,Rehmannia glutinosa(Gaertn)Libosch, Ligusticum chuanxiong Hort,Paeonia lactiflora Pall).We detected the activities and the change of Acetylcholinesterase(AchE),Superoxide Dismutase(SOD), Glutathion peroxidase(GSH-Px)and Brain-derived nerve factor(BDNF)in the brain tissue of imitative Vascular Dementia rat.We also observed the influence of constitutes of Siwutang on these enzymes and cytokine,and analyzed the efficacy of every constitute on VaD.Method:The imitative Vascular Dementia rat model was established by repeatedly occluding the common carotid artery of the rat in combination with an abdominal injection of Sodium Nitroprusside.We divided 70 rats randomly into 7 groups,which were pseudo-operation group,model group,positive control group, group and Angelica sinensis and Ligusticum chuanxiong Hort group,Ligusticum chuanxiong Hort group,Rehmannia glutinosa(Gaertn)Libosch and Paeonia lactiflora Pall group,Rehmannia glutinosa(Gaertn)Libosch,Paeonia lactiflora Pall and Angelica sinensis group respectively.The therapy groups were treated with different decoction of the herbs after operation.On 14^thdays,we took out the brain quickly,then made 10%tissue homogenate,centrifugalized it under 4℃, measured the activity of AchE,SOD and GHS-Px in the supernate.The content of BDNF in the supernate was measured with ELISA.Conclusion:The results shows that under different therapy,AchE,GSH-Px,SOD and BDNF in each group's brain tissue of imitative VaD rat have a clear change, but the extent of this change has no notable discrepancy among these groups,so we can draw a conclusion that the efficacy of Siwutang on VaD is multiple targets, it is a conjunct effect so that maybe the key constitutes don't exist.;Rehmannia glutinosa(Gaertn)Libosch and Paeonia lactiflora Pall group,Rehmannia glutinosa(Gaertn)Libosch,Paeonia lactiflora Pall and Angelica sinensis group have better efficacy on strengthening the activity of GSH-Px,this illuminated that Rehmannia glutinosa(Gaertn)Libosch or(and)Paeonia lactiflora Pall can strengthen the activity of GSH-Px in brain tissue of imitative VaD rat,which is good for mitigating the hurt of free radical on brain tissue.
