Objective: The purpose of this research is to explore the acting mechanism of Siwutang (Decoction of four herbs, Angelica simnsis (Oliv.) Diels, Rehmannia glutinosa (Gaertn) Libosch, Ligusticum chuanxiong Hort, Paeonia lactiflora Pall) and neuroprotection of cytokine in treating Vascular dementia. We detected the activities of Acetylcholinesterase (AchE), Superoxide Dismutase (SOD) and GSH-Px in the brain tissue of imitative Vascular Dementia rat. We also detected the contents of Epidermal growth factor(EGF) and Brain-derived nerve factor(BDNF) in rat brain tissue with enzyme-linked immuno-sorbent assay(ELISA),and observed the influence of Siwutang on these enzymes and cytokines. Thereby we can analyze the preventive and Therapeutic efficacy of Siwutang on Vad, It will lay foundation for prevention and treatment of VaD.Method: the imitative Vascular Dementia rat model was established by repeatedly occluding the common carotid artery of the rat in combination with an abdominal injection of Sodium Nitroprusside. We divided 50 rats randomly into 5 group, which were pseudo-operation group, model group, positive control group, low dose group and high dose group respectively. Eleven days after ischemic-reperfusion, intelligence tests were carried out. On 14th days, we took out the brain tissue quickly, then made 10% tissue homogenate, centrifugalized it under 4℃,measured the activity of AchE, SOD and GHS-Px in the supernate. The content of EGF and BDNF in the supernate were measured with ELISA.Result: The results showed that Siwutang improved learning and memorial ability; improved the activity of SOD and GHS-Px, and lowered the activity...
