Study On The Expression Of MUC2,MUC5AC,TFF1,TFF3 In The Ulcerative Colitis Patients With Large Intestine Damp-Heat Syndrome

Objective:To reveal the regularity of the expression of MUC2,MUC5AC,TFF1,TFF3 in the ulcerative colitis patients with Syndrome of large intestine Damp-Heat,Syndrome of spleen-stomach Qi deficianey and healthy persons,to approach the biological basis of the pathogenesy of Syndrome of large intestine Damp-Heat and the relationship between the different Syndrome Differentiation-Types using modern medicine targets at the molecular level,and to provide a theoretical basis for clinical diagnosis and treatment.Methods:74 cases of ulcerative colitis were selected.There were 48 cases of Syndrome of large intestine Damp-Heat(LIDH),26 cases of Syndrome of spleen—stomach Qi deficiency(SSQD),and 10 common people were selected as control group.The MUC2,MUC5AC,TFF1,TFF3 of intestinal epithelium were obtained by immunohistochemical technique.Results:1.The expression of MUC2 in Ulcerative colitis in LIDH group and SSQD group were lower than that in the normal control group,but the difference was not significant(P>0.05). The MUC2 expression of strong masculine rate in LIDH group(41.67%)was slightly higher than that in SSQD group(38.46%),without significant difference(P>0.05).2.The expression of MUC5AC in Ulcerative colitis in LIDH group and SSQD group were higher than that in the normal control group,the difference between LIDH group and the normal control group was significant(P<0.05),but the difference between LIDH group and the normal control group was not significant(P>0.05),and the difference between LIDH group and SSQD group was not significant(P>0.05).3.The expression of TFF1 in Ulcerative colitis with LIDH was 12.5%,higher than that in the normal control group of 10.0%,but they were not significant(P>0.05),There was no expression of TFF1 in SSQD group,and the difference between SSQD group and the normal control group was not significant(P>0.05),the difference between SSQD group and LIDH group was not significant(P>0.05).4.The expression of TFF3 in Ulcerative colitis in LIDH and SSQD groups were lower than that in the normal control group,and the difference was significant(P<0.05).The strong masculine expression of TFF3 in Ulcerative colitis with LIDH was higher than that in SSQD groups,but it was not significant difference between this two groups(P>0.05).Conclusion:1.The expression of MUC2 in Ulcerative colitis with LIDH and SSQD groups were not different from but both lower than that in the normal control group,which means the expression of MUC2 in Ulcerative Colitis is lower than in the normal control group.There was not significant difference between the LIDH group and SSQD group,but the expression of strong masculine rate of MUC2 in LIDH group was slightly higher than that in SSQD group,which may indicate a diminishing trend of the expression of MUC2 in this two Syndrome Differentiation-Types,which might prompt the direction of the development of the disease,and could provide a valuable signal for the disease prognosis and treatment.2.The expression of MUC5AC in Ulcerative colitis in LIDH and SSQD groups were higher than that in the normal control group,the difference between LIDH group and the normal control group was significant,which suggested that the expression of MUC5AC played a part in the occurrence and development of UC with LIDH group,but so far,the reason was still unfound.3.The expression of TFF1 in Ulcerative colitis with LIDH was 12.5%,but there was no expression of TFF1 in SSQD group,which could suggest that the healthy qi of LIDH group is deficient,and the defense function of their intestinal mucosa decline.4.The expression of TFF3 in Ulcerative colitis in LIDH and SSQD groups were lower than that in the normal control group,and their difference was significant.And the expression of TFF3 in Ulcerative colitis in LIDH group is slightly higher than that in the SSQD group,which suggested that the defense function of intestinal mucosa in LIDH group is lower than that in SSQD group.It might prompt the direction of the development of the disease.Meanwhile,it was reported that TFF3 could cure the impairment of gastrointestinal mucosa,which could provide a new way for the treatment of ulcerative colitis.
