ObjectiveTo study the roles of CD147, Monocarboxylate Transporter 1 (MCT1) and Monocarboxylate Transporter 4 (MCT4) in the pathology of Squamous cell carcinoma (SCC) and its clinical significance.MethodsFormalin-fixed paraffin-embedded skin skin biopsy specimen blocks from SCC patients and normal controls were used in this study, including 12 cases of poorly differentiated SCC (PSCC), 18 cases of moderately differentiated SCC(MSCC), 39 cases of highly differentiated SCC (HSCC), and 20 cases of normal skin controls (NS). Immunohistochemical method was applied to detect the expression of CD147, MCT1 and MCT4 in 89 specimens. An immunoreactivity intensity distribution index (IRIDI) was calculated to express the proportion of the immunoreactive cells as well as the staining intensity in those cells. The positive rate of CD147, MCT1 and MCT4 were calculated by an immunoreactivity intensity distribution index (IRIDI).Results1. CD147, MCT1 and MCT4 staining were observed in every group.2. The expression level of CD147 and MCT4 in SCC was remarkably higher than those in the normal skin (all P<0.05), and associated with the classification of SCC.3. The expression level of MCT4 in SCC was remarkably higher than that in the normal skin, and its expression in PSCC was remarkably higher than that in MSCC and HSCC (all P<0.05). Its expression between HSCC and MSCC has no statistical difference (P>0.05).4. The expression level of CD147 was positively correlated with the expression level of MCT1 in SCC (P < 0.05).5. The expression level of CD147 was positively correlated with the expression level of MCT4 in SCC (P < 0.05).6. CD147 and MCT1 are mainly distributed in the cell membrane, cytoplasm is also a small amount of the distribution; MCT4 is mainly distributed in cytoplasm and mainly in the distribution of the cytoplasm region near the nucleus of the tumor cell.Conclusions1. The elevated expression of CD147 and MCT4 of SCC could be associated with the classification of SCC.2. CD147, MCT1 and MCT4 might play important roles in the process of the cutanous cancer.
