Cyclocarya paliurus (Batal.) Iljinskaja is a precoius plant resource in China, it has many biological activities for it contains many kinds of bioactive components. This research was conducted to investigate the hypolipemic effect of Cyclocarya paliurus (Batal.) Iljinskaja polysaccharide in the induced-hyperlipoidemia mice, the total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL-C), high density lipoprotein (HDL-C), free fatty acid (FFA), and lipase (LPS) levels in the serum were detected, and the effect of the polysaccharide on the pathological changes in the liver of the mice were observed; the effect of polymorphism of fatty acid synthase (FAS) gene on the blood-fat traits was invesigated by PCR-RFLP, meanwhile the mRNA and protein expression of the gene in the different tissues were detected by RT-PCR and western blot respectively. The results showed that:(1) Cyclocarya paliurus (Batal.) Iljinskaja polysaccharide has significant hypolipemic effect. The level of serum TC,TG,LDL-C,LPS in mice fed with polysaccharide could be remarkably reduced (P<0.01), and the HDL level could be remarkably increased (P<0.01), and among which the effect of high does(400 mg/kg/d) was the best.(2) Cyclocarya paliurus (Batal.) Iljinskaja polysaccharide can significant improve the hepatic fatty change of experimental hyperlipoidemia mice, and can reduce the hepatocytes fat vacuoles obviously.(3) Different FAS genotypes showed different influence on blood-fat traits and some organ performance of mice. Different FAS genotypes can dramatically affected the heart, adipose weight of mice, also the ratio of heart to body weight and adipose to body weight.(4) Cyclocarya paliurus (Batal) Iljinskaja polysaccharide could remarkable restrain the expression of FAS mRNA in some organ performance of the hyperlipoidemia mice.(5) Cyclocarya paliurus (Batal) Iljinskaja polysaccharide could remarkable restrain the expression of FAS protein in liver of the hyperlipoidemia mice.
