The Effects Of LWDHW-Containing Serum On Expression Of Type 1 And Type 2 Collagen In Rabbit's Intervertebral Discs

Objeetive:To observe the effect of serum containing LWDHW on type I and typeⅡcollagen of the intervertebral discs of rabbit which were injuryed by TNF-a.To discussion whether LWDHW has protective effects of ECM of Intervertebral discs?Method:Intervertebral discs which were cut from T12-L5 of 15 rabbits by operation were randomly divided into five groups including the sham group in 1st day(group A), the sham group in 14th day (group B), the TNF-a group(group C),the serum containing LWDHW(group D) and the TNF-a+serum containing LWDHW (group E). nuelcuspulposus and fibrous rings cut from T12-L2 were observed by optical microscope; nuelcuspulposus and fibrous rings cut from L2-L5 were detected by RT-PCR detection and western blot test.The samples in group A were observed and detected right after the grouping.Establish the vitro culture system of the intervertebral discs,add the TNF-α(5 ng/mL) into the group C,add the serum containing LWDHW(10%) into the group D, add the TNF-a(5 ng/mL) and the serum containing LWDHW(10%) into the group E.The intervertebral discs in the left four groups were observed by optical microscope and detected by RT-PCR detection and western blot test in the 14th day of the experiment.Result:1. observed by optical microscope:the number of cells and collagen content in group A, B, E and C were progressively less. 2. RT-PCR detection and western blot test:compared to group A,there were more typeⅠcollagen and less typeⅡcollagen in group B (P<0.01).The contents of typeⅠcollagen in group C were more than that in group B, the downward tendency of the typeⅡcollagen was distinct than that in group B(P<0.01). The contents of typeⅠcollagen in group E were more than that in group C, the downward tendency of the typeⅡcollagen was mild than that in group C (P<0.01).Conclusion:With the effect of protecting typeⅡcollagen from the inhibited effect of TNF-α,the LWDHW can protect the ECM of Intervertebral discs and provide a experimental reference for treating discogenic back pain by the method of invigorating kidney.
