The Effect Of Yishen Capsule On SOCS-3and Collagen TypeⅠ、Ⅳ In Diabetic Nephropathy Rats

Objective:To observe the effect of Yishen capsule on SOCS-3、collagen type I and IV in Diabetic nephropathy rats and to investigate the mechanism Of Yishen Capsule to delay diabetic nephropathy fibrosis.Methods:The rat models of diabetic nephropathy were established by intraperitoneal injection of STZ (Streptozotocin60mg/kg). Then the rats were randomly divided into three groups:DN group (only distilled water by gavage), Y group (Yishen capsule625mg-kg-1-d-1by gavage), L group (Losartan30mg·kg-1·d-1by gavage). The additional set of healthy rats were given no STZ were used as control group (N group). Each group consisted of12rats. By12weeks, the body weight,24h urinary albumin excretion, serum creatinine (Scr) and blood urea nitrogen (BUN) level were measured in each rat. Then, renal tissue samples were observed under optical microscope. Expression of SOCS-3、Collagen type I、IV in renal tissue samples were detected by immunohistochemistry. Results:Compared with N group, DN group had significant elevated24h urinary albumin, Scr and BUN (P<0.05), the pathological changes in the renal tissue samples in DN group were also obvious. DN model rats, renal tissue mesangial proliferation, glomerular hypertrophy, glomerular capillary basement membrane thickening, part of renal tubule happen vacuolar degeneration, renal tubular atrophy, Part of the renal capsule cavity expansion, balloon conglutination; The pathological changes in the renal tissue samples were much relieved in the treated groups, the treated groups:the renal tissue mesangial cells and mesangial proliferation, glomerular hypertrophy matrix reduce significantly reduced, a few renal tubule expansion. Masson statining:blue collagen fiber were distributed in DN group; PAS statining:a lot of red particles were detected in DN group. Compared with DN group, the treated groups had significant lower24h urinary albumin, Scr and BUN (P<0.05). The expression level of SOCS3in treated group was significant higher than that in DN group, but the expression level of collagen type Ⅰ、IV were lower than that in DN group (P<0.05).Conclusion:Yishen capsule can reduce urinary protein and improve kidney function, at last to delay diabetic nephropathy renal fibrosis, its mechanism is Yishen capsule may through the up-regulation of SOCS3expression to inhibit the expression of Collagen type I、IV, thus reduce the degree of the DN rat glomerular sclerosis and renal tubule interstitial fibrosis, then delay the progress of diabetic nephropathy.
