Objectives:Though the observation of the expression of YiShenTong LuoDecotion effects on Membranons Nephropathy (MN) model rats and its impact on the rats with 24-hour urine protein, blood biochemistry results, the expression of heparanase and transforming growth factor in kidney tissues, and compared with Benazepril group, try to further confirm that the Decoction's effect and mechanism of MN from an experimental point of view, explore and research new treatment methods and effective drugs, in order to provide an experimental basis for clinical and broaden the therapeutic areas of MN.Methods:48 eight-week-old healthy male SD rats were randomly divided into four groups of 12, respectively, the normal group (NG), model group (MG),YiShenTongLuo group (ZG) and Benazepril group (BG).Using the Border method to induce models rats of MN.1week after the pre-immunization,give MG,BG,and ZG 3 times a week via tail vein injection of cationic bovine serum albumin (C-BSA) to copy MN rats,the NG injected the same dose of water, continuous injection for 4 weeks,after the success of modeling,give BG Benazepril ydrochloride 10mg/kg/d gavage, give ZG Yishen Tongluo Fang decoction 5ml / d gavage, NG and MG were fed the same amount of saline, the rats freely eating, drinking water.Rats in each group before making model,after modeling (at the end of the 5 week) and after treatment (at the end of the 9 week)metabolism in rat cage to retain an 24-hour urine, a 24-hour urine protein quantitatively. Rats in each group were then sacrificed, abdominal aortic blood, were detected in serum total protein (TP), albumin (ALB), total cholesterol (TC), triglyceride (TG), blood urea nitrogen (BUN), creatinine (Scr),and specimens from kidney tissue, respectively for the light microscopy,immunofluorescence and electron microscopic observation of glomerular basement membrane and foot process changes, using immunohistochemical staining to check the expression of Hpa and TGF-β1 in the kidney tissue ,and the pathological image analysis system for semi-quantitative analysis.Results:1.24h urinary protein in each group of rats 4 weeks after the tail vein injection of C-BSA,MG (26.86±3.66), BG (26.20±1.58) and ZG (25.60±2.52) were massive proteinuria in rats with NG (6.40±1.21),tatistically significant (P <0.05); administration after 4 weeks, ZG (10.18±1.15) and BG (14.71±2.42) 24-hour urine protein quantity were significantly reduced,compared with MG (25.03±7.28),tatistically significance (P <0.05);ZG (10.18±1.15) compare with BG (14.71±2.42),statistically significant (P <0.05). compared with before treatment (25.60±2.52), ZG after treatment (10.18±1.15) p <0.05, statistically significant, compared with before treatment (26.20±1.58),BG after treatment (14.71±2.42),p<0.05, there is statistical significance.2.The results of blood biochemistryAfter treatment,the serum TP,ALB of ZG,BG were elevated in diffient degrees,compared with MG(60.67±5.09,29.59±3.01),statistical significance (P <0.05), ZG (74.19±3.21,38.28±2.46) compared with BG (68.16±3.22,32.95±1.45),statistical significance (P <0.05);the serum TC, TG of ZG,BG were reduced in diffient degrees, compared with MG(4.82±0.71,0.67±0.18),statistical significance (P <0.05), ZG (2.76±0.10,0.31±0.03) compared with BG (3.22±0.39,0.39±0.03) statistical significance (P <0.05);each group serum BUN, Scr compared with NG, no statistical significance (P> 0.05).3.ght and electron microscopy and immunofluorescenceLight microscope show:NG have no obvious abnormalities glomerular structure, MG glomerular volume increased significantly, the ball is inside the cells increased markedly thickened capillary wall, glomerular surrounding a large number of inflammatory cell infiltration, glomerular lobulation obvious, renal proximal tubule edema,Masson staining showed diffuse glomerular basement membrane thickening, segmental nail sudden formation of a large number of renal tubular epithelial cell degeneration and granular degeneration, can see a small amount of renal interstitial lymphoid and mononuclear cell infiltration, fibrosis, wall thickening of small arteries. ZG glomerular structural change was not obvious, BG glomerular volume increased slightly, the ball inside the cells increased, capillary wall thickening was not obvious.Electron microscope show:NG have a very small amount of electron-dense material deposition, there is obvious podocyte foot processes, MG can be seen in the majority of electron-dense glomerular basement membrane deposition, electron dense material surrounding a large number of basement membrane hyperplasia, thin film matrix hyperplasia, diffuse foot process fusion, ZG within a few electron-dense glomerular basement membrane deposition, some diffuse foot process fusion, BG electron-dense material within the glomerular basement membrane deposition, electron-dense material around the basement membrane hyperplasia, Thin film matrix proliferation, some diffuse foot process fusion.MG visible immunofluorescence showed fine granular IgG and C3 along the glomerular capillary wall and some of the mesangial deposition. NG performance is not obvious, ZG shows mild IgG and C3 deposition, BG can see a small amount of IgG and C3 fine granular deposits along the glomerular capillary wall.4.mmunohistochemical stainingAfter treatment, the Hpa (LOD values) nd TGF-β1 positive rate (%) of ZG and BG in kidney tissue were reduced in diffient degrees, compared with the MG (85.06±5.99,55.33±6.92),statistical significance (P < 0.05), ZG (23.42±3.78,26.97±5.74) compared with BG (54.53±9.73,35.75±5.63),statistical significance (P <0.05).Results:1 Spleen and kidney, promoting blood circulation meridians" is a method of treatment proposed by my teacher the basis characteristics of this virtual MN real incidence.Experimental studies confirmed Yishen Tongluo Decotion is reflected in it.2. The experimental results show that Yishen Tongluo Decotion can reduce the content of MN rats 24h proteinuria, improve TP and ALB, lower TC and TG,adjust the expression of Hpa and TGF-β1.3 Yishen Tongluo Decotion can reduce the MN rats immune complexes deposition in the GBM,reduce the thickening of GBM,and promote MN histopathologic damage repair. More effective than benazepril hydrochloride.4. The mechanism of Yishen Tongluo Decotion in the treatment of MN needs further deepened.
