The Lepidium latifolium L.,it’s content of flavonoids was high in lonicera lonicerata,and flavonoids had a variety of biology effects.The flavonoids content in the crude flavonoids extracted from lonicera grandis by traditional extraction method was low.In this study,we selected the broadleaf cultivar from Qinghai Province,extracted and purified the flavonoids,and studied the fingerprints of L.latifolium.L.in different regions of Qinghai Province in order to comprehensively utilize the broad-leaved lone development provides a reference.(1)Using ultrasonic assisted extraction method and ethanol as extraction solvent,the effects of ethanol concentration,liquid-to-liquid ratio,extraction temperature and extraction time on the extraction content were investigated by single factor test and response surface test.The optimized extraction conditions were as follows: ethanol concentration of 75%,extraction time of 89 min,liquid-to-liquid ratio of 22.5 mL/g,and extraction temperature of 79℃,the flavonoid content of L.latifolium.L.was up to 36.5mg/g.(2)By compared the static adsorption,analytical parameters and partial dynamic adsorption parameters of D301,D201,D-101-1,X-5 and AB-8 macroporous resins,to determined that AB-8 macroporous resin was determined to be the most suitable for purification of flavonoids in the leaved cabbage.Purification test of flavonoids from L.latifolium.L.The static single factor experiment was carried out on the purified flavonoids of L.latifolium.L.,and the optimum adsorption concentration of the test solution is 4.34 mg/mL,the optimum pH is 6,the optimal elution concentration is 70% ethanol,and the static desorption equilibrium time is 2 h.Through the dynamic single factor test on the purification of total flavonoids from L.latifolium.L.,it is determined that 70 mL of flavonoid solution is selected as the best loading amount of 4.34 mg/mL,and the elution volume was 80 mL.Under the optimal purification conditions,the purity of the flavonoids of L.latifolium.L.was 38.9%.(3)HPLC fingerprint patterns of different producing areas were established through 10 batches of L.latifolium.L.samples,and 12 common peaks were confirmed.The no.12 peak was identified as Rutin.The common chromatographic peak similarity of 10 batches of L.latifolium.L.HPLC fingerprints was greater than 0.90,showing strongcharacteristics and specificity(4)Comparing the chemical constituents in the volatile oil extracts of L.latifolium.L.chinensis in five different regions of Qinghai Province,there are some differences in the composition of hair oil extracts,but the main components are mainly isothiocyanates,especially the allyl isothiocyanate content is the highest.
