This paper studies on the extraction technology, separation technology,amplificated industrialized production and the qualitative and quantitative analysis of chlorogenic acid in Eucommia leaf to obtain the better theoretical basis and experimental result in Industrialized production.1.According to single-factor experiment and orthogonal experiments, it was indicated that the optimum extraction condition of chlorogenic acid from eucommia leaf by ultrasonic wave and ethanol was ethanol concentration 75%, solid-liquid ratio 1:10 and 30 min of extraction for twice. The extraction rate of ultrasonic extraction was higher than that it by hot reflux 8.5%. Compared with the hot reflux, the ultrasonic extraction saved more time and more cost of manufacture.2. Macroporous adsorption resin screening experiment shows that the ideal purification concentration chlorogenic acid in eucommia adsorption materials is LSA-21 macroporous resin. Dynamic adsorption experiment's conclusions are as follows:In pH 3-4, selected liquid concentration for 6.67 mg/ml, sample weight in 2BV, sample flow control in 1.0-1.5 BV/h, and 1.5-2.0 BV/h with no ion water to nearly colorless, using 6BV 50%alcohol for elution and the flow should be controled in 2.0 to 2.5 BV/h,Then there is 93.92%chlorogenic acid desorption and the purity is 30.24%;If using 6BV 70%ethanol for elution and the flow in 2.0 to 2.5 BV/h, there is 95.12%chlorogenic acid desorption and the purity is 22.97%. Compared with no resin purification, the purity of chlorogenic acid has increased 6 times and 4.5 times respectively,and the result shows that the concentration of ethanol can obtain different content on the eluate products, according to the market requirements,different processes can be choosen to meet them.3. The result of Eucommla ulmoides chlorogenic acid extraction and separation in the industrial production amplification is:We have the content of chlorogenic acid is 30.57%and the average productivity is 3.52% by using the 50% alcohol as flow.If using 70% alcohol as flow,the average content of chlorogenic acid is 23.48% and the average productivity is 4.62%. We can get high level of chlorogenic acid products through further purification.4.The measures of analysis and detection of chlorogenic acid are as follows:paper chromatography, the TLC, paper chromatography (the TLC)-ultraviolet spectrophotometry and HPLC. Paper chromatography, the TLC because of its simple, convenient and do not need expensive instruments, which can be as chlorogenic acid, the method of qualitative tests. From the resolution, rapid aspects thin layer chromatography better than paper chromatography, thin layer chromatography in enterprise production is the preferred method of on-line detection; Paper chromatography ultraviolet visible photometric, HPLC due to its high accuracy can be used as a quantitative detection methods,and from the resolution, rapid and accuracy aspects HPLC significantly better than paper chromatography ultraviolet,so HPLC is the preferred method of accurate detection in enterprise production.
