In this investigation,root and xylem of Eucommia ulmoides Oliv.were extracted with 95%EtOH and then sequentially fractionated with a series of solvents with increasing polarity to yield soluble fraction.The content of total flavonoids and total phenolics of each fractions were measured,and the antioxidant,anti-inflammatory and anticancer activities were analyzed.Moreover,the extractions of polyphenols of E.ulmoides root and xylem were optimized by response surface methodology respectively,and the data was analyzed and forecasted by Design Expert.The results could provide a certain theoretical foundation for the exploitation and utilization of E.ulmoides root and xylem.?1?Total flavonoid content?TFC?and total phenolic content?TPC?of E.ulmoides root and xylem were measured by NaNO2-Al?NO3?3-NaOH coloration system and Folin-Ciocalteu reagent.Results showed that TFC and TPC were the highest in EtOAc fraction from xylem?290.3±6.26 and 215±23.27 mg/g,respectively?,and the lowest in H2O fraction from xylem?32.15±0.28 and 28.66± 1.39 mg/g,respectively?.?2?The antioxidant ability of extracts of E.ulmoides root and xylem was evaluated by DPPH free radical scavenging activity and reducing power capacity.And results showed that EtOAc fraction of E.ulmoides root and xylem exhibited more efficient DPPH and reduced more Fe3+ than the others,and the H2O fraction was the weakest.In the study of anti-inflammatory activity,n-hexane,CH2Cl2 and EtOAc fractions showed significant inhibition for the inflammation induced by LPS;However,only n-hexane fraction of E.ulmoides root had the ability to reduce the cell viability of HT-29 in the study of anticancer effect,and the inhibition ratio was up to 96.06±0.10%at the concentration of 100 ?g/ml.?3?The extraction of polyphenols of E.ulmoides root and xylem were studied with the method of ultrasonic assistant extraction,and the extraction conditions were optimized by combining single factor and response surface methodology.In order to explore the effect of ultrasound on the impact of the extraction in this study,homothermal ultrasonic extraction was used to keep the temperature steady when sonicating by setting up a water cycle between ultrasonic equipment and thermostat water bath.The results of response surface methodology were showed as followed:i)The optimal conditions of polyphenols extraction of E.ulmoides root were extraction temperature 66?,preimpregnation time 35 min,extraction time 41 min,concentration of EtOH 62%,ratio of liquid to material 30 ml/g,and the yield of polyphenols of E.ulmoides root was 16.04 mg/g,the effect was decreasing by the order of extraction temperature,preimpregnation time,extraction time,concentration of ethanol,ratio of liquid to material;ii)The optimal conditions of polyphenols extraction of E.ulmoides xylem were extraction temperature 63?,extraction time 52 min,concentration of EtOH 45%,ratio of liquid to material 37 ml/g,preimpregnation time 41 min,extraction times 3 time,and the yield of polyphenols of E.ulmoides xylem was 6.21 mg/g,the effect was decreasing by the order of extraction times,concentration of ethanol,ratio of liquid to material,extraction time,extraction temperature,preimpregnation time.
