Objective:In order to clarify the chemical constituents of SQWWZT as well as its absorbed constituents,prototypes and metabolites after oral administration of SQWWZT,a rapid and sensitive UPLC-HDMS/MS and method was successfu lly established,which also showed available for analyzing the constituents of Chinese medical formula in vitro and in vivo and helping to further reveal th e material foundation of SQWWZT basing on Serum Pharmacochemistry of T CMMethods:Guidied by Serum Pharmacochemistry of TCM,the constituents of SQW WZT and constituents absorbed into blood after oral administration of SQW WZT were detected and characterized by means of UPLC-HDMS/MS technol ogy.The chromatograms acquired by UPLC-MS/MS were processed through t he intuitionistic comparation and the MassLynx data processing technology r espectively.Results:1 Analysis of the constituents in vitroBy the method of analyzing SQWWZT constituents in vitro,an injection run of the SQWWZT sample was finished in only 17min,and excellent chrom atographic separation and sensitive detection were obtained.As a result,101 chemical compositions were found in SQWWZT objectively.Among them,92 compounds were identified,which mainly included Lignans?Flavonoides?A lkaloids?Saponins?Organic acids et al,such as Deoxyschisandrin?Schisan drin B?Schizandrin?Schisandrol B?Chicanine?Schisandron?Codonopsin e?Tangshenoside I?Tangshenoside V?Astragaloside I?Astragaloside I V?Spinosin?Ononin?Claycosin?Jujuboside A?Jujuboside B et al.2 Analysis of the constituents in vivoOn account of the developed method of fingerprint analysis of the SQW WZT,an injection run of the sample of dosed rat plasma was finished in 17.0 min.Through the intuitionist comparisons of the three chromatograms and ap plications of multiple data processing technology,41 compounds were detecte d and characterized in rat plasma.A total of 35 prototype components and 6 metabolites were identified and characterized in rat plasma after oral admini stration of SQWWZT,including lignans,teroidal saponins,flavonoids and alka loids et al.ConclusionAccording to Serum Pharmacochemistry of TCM,the wide use of UPLC-HDMS/MS and various data processing technology contribute to global analy sis of the constituents of SQWWZT in vitro and in vivo.As a result,41 compo nents in rat serum after oral administration of SQWWZT are characterized.w hich could be the effective substances of SQWWZT for treating insomnia.Th e research may play a key role in clarifying the material basis involved in th erapeutic effects of SQWWZT and also providing an example for further stud y on constituents of TCM in vitro and in vivo.
