Four Needle Comparative Study Of The Analgesic Mechanism Of The Hypothalamus In Rats With Adjuvant Arthritis

Objective: Comparative research was designed to prove the analgesia efforts of electro-acpuncture, filiform needle acpuncture, blood-pricking therapy, medicine injection therapy and analgesia mechanism in hypothalamic.Methods: The experiment was used adjuvant arthritic(AA) rats as the model of inflammatory-pain, selected " KunLun " (UB-60),adopted electro-acpuncture, filiform needle acpuncture, blood-pricking therapy and medicine injection therapy,used observed methods of radiommunoassay, in situ-hybridization histochemistry, and observed targets of pain threshold, swelling degree in inflamed area, that of P-EP, ACTH, the genie expression of POMCmRNA in hypothalamus.Results: Four kinds of therapies all raise pain threshold, especially in electro-acpuncture and medicine injection therapy; release swelling degree in inflamed area, not much difference. Electro-acpuncture and filiform needle acpuncture both can raise the contents of P-EP in hypothalamus, espscially in electrotherapy, not influence of medicine injection therapy and blood-pricking therapy; medicine injection raise the contents of ACTH in hypothalamus, not include of the others. Electro-acpuncture and filiform needle acpuncture raise genie expression of POMCmRNA, not influence of medicine and blood-pricking.Conclusion: The four methods all observe the effects of analgesia to inflammatory-pain, especially in electro-acupuncture and medicine injection therapy.In hypothalamus, electro-acpuncture and filiform needle acpuncture maybe observe the effects of analgesia by raising the content of P-EP in hypothalamus and the genie expression of POMCmRNA. By raising the contents of ACTH in hypothalamus, medicine injection therapy maybe have the effort of analgesia, blood-pricking therapy maybe have the effort of analgesia besides p-EP and ACTH in hypothalamus.
