This dissertation is composed of two sections. The first is about studies on chemical constituents of selaginellaceae Selaginella tarmariscina (Beauv.) spring. The second involves studies on quality standard of selaginellaceae Selaginella tarmariscina (Beauv.) spring.Juanbai,The dried whole plants of Selaginella tamariscina (Beauv.) Spring,which belongs to Selaginellaceae .which is also named Changshengbusicao, jiusihuanhuncao, shilianhua, wannianqing etc. which and Selaginella pulvlnata (Hook et Grev.) maxim. are always accepted as Juanbai national standard material medicine in China Pharmacopeia (I). It is a traditional medicine and recorded as best medicine in Benjing Purgent. Extracts of Selaginella tamariscina have been reported to remove blood stasis,promote blood circulation and arresting, and mostly used to cure abdominal pain, amenorrhea, mass in abdomen,spitting blood, bloody urine and hematochezia etc. In addition, Modern pharmacological experiment and clinical studies reveals it has a series of functions such as lowering blood glucose levels , facilitating the repair of pancreatic islet B cells injured by alloxan and reducing the producion of proinflammatory cytokines,interleukin-1 and tumor necrosis factor in human mesangial cells. In addition, crude extracts of Selaginella tamariscina have other functions.for example, anticancer, anti-inflammation, antivirus, easing pain, lowering blood pressure, enhancing human immunologic function,and so on.9 compounds were isolated from ethanol-extracts of Juanbai.6 of them were identified on the basis of spectral and chemical data, including UV, IR, 1H-NMR, 13C-NMR. they are: amentoflavone, D-mannitol,coffeic acid,vanillic acid,ferulic acid,β-Sitosterol.while different qualities would were established by HPLC method.change slightly with different change sharply. So we think that quantitative of amentoflavone could be as Selaginella tarmariscina (Beauv.)Spring a quality control reference of Selaginellaceae. Firstly,the preparation procedure of reference compound was studied;Secondly, To determine the content of amentoflavone in S. tamariscina with RP-HPLC method, Agilent zorbax C18 analytical column (250mm×4.6mm, 5μm) was used at room temperature, The mobile phase consisted of methanol (A) and 0.1% formic acid in water (B) using a gradient program, the gradient elution had the following profile: 10% (A) in 0-10 min, 1060% (A) in 10-20 min, 6080% (A) in 20-50 min. The flow rate was 0.8 mL/min. DAD detector was set at 330 nm for acquiring chromatograms. The injection volume was 10μL. Amentoflavone and showed, 0.06112.444μg, a fine linear relationship with the peak areas ( r = 0.9999,n = 6 ). The average recovery of this procedure was 99.07%, with RSD equal to 1.79% ( n = 6 ). With a good repetition, this procedure is easy and accurate, and can be used in the analysis of the quality of amentoflavone of S. tamariscina.The fingerprint chromatogram of entire herb constituents of Selaginellaceae selaginella tarmariscina (Beauv.) Spring was developed by HPLC method for the first time. High-performance liquid chromatography paired with photodiode-array detector was analyzed as a method based on the consistent chromatographic features among 10 batches of authentic samples. The HPLC fingerprint of S. tamariscina has been established for the first time. According to the chromatographic condition, a good fingerprint of S. tamariscina was obtained. All chromatographic peaks showed good resolution in the liquid chromatogram. This work has not been done systematically by other scholars. The unique properties of this HPLC-DAD fingerprint were successfully applied to analyze and differentiate samples from different regions. The results showed that these variations will give rise to differences in identities and indicate that a comprehensive quality evaluation of Chinese herb S. tamariscina, which is critical to assess and represent its overall quality.
