PartⅠThe study of asthenospermia animal model induced by Ouabain in SD ratsObjective : To investigate the pathological mechanism of Ouabain-induced asthenospermia, this research estabished asthenosrermia animal model in SD rats by injecting Ouabain.Methods:Forty adult male SD rats were randomly divided into control group (N=10, saline 10mg/kg/d), low dose experimental group (N=15, Ouabain 12.5ug/kg/d) and high dose experimental group (N=15, Ouabain 25ug / kg / d). After intraperitoneal injecting for 30 days, we had observed each group for their epididymal sperm motility, activate rate and the possible toxicity of reproductive system and the body.Results:Compared with control group(73.33%) ,the high and low dose group's epididymal sperm motility is respectively 48.27% and 44.73% ; And epididymal sperm activate rate respectively decrease to 52.26% and 49.95% compared with control group (78.70%); The differences among the epididymal sperm motility of high dose group,low dose group and contorl group were significant(p<0.01);The differences among the sperm activate rate of these three group were significant(p <0.01) too; Ouabain on reproductive organs and the body of important organs in rats did not show obvious toxicity in test dose range.Conclusion:Asthenospermia SD rats model without significant reproductive and systemic toxicity were successfully established by intraperitoneal injecting Ouabain for 30 days .PartⅡThe exploratory study on the function of secretion Ouabain by Leydig cell in mouseObjective: Detected the function of Leydig cells secreting Ouabain intervented by hCG and FSH in mouse to explore the relationship of Ouabain and asthenospermia. Methods:Incubated Leydig cells in vitro, intervened by different concentrations of hCG (0U, 4U, 8U, 16U, 32U, 64U, 100U, 200U) and FSH (0,5,10,20,50,100 mU / ml), collected the supernatant ,and detected the level of Ouabain in supernatant by ELISA.Results:Ouabain can be detected in supernatant of Leydig cells intervented by hCG ,and the level of ouabain is 49.82pg/ml when concentration of hCG is 4U, increases to 60.70 pg / ml when hCG is 8U,and up to 259.13 pg / ml when hCG is 200U. However,Ouabain can't be detected in supernatant intervented by FSH.Conclusion: Leydig cells can synthesize Ouabain,and, the level of Ouabain increases with elevation the concentration of hCG. But the effect of FSH on Leydig cells has not seen in test dose range.
