| Objective To investigate the expression level of vascular endothelial growth factor-C(VEGF-C), hepatocyte growth factor(HGF) and its receptor c-Met in cervical carcinoma , also test the levels of serum HGF and VEGF-C. To analyze the clinical significance of VEGF-C, HGF and c-Met and their relationships.Methods (1) The mRNA levels of VEGF-C,HGF and c-Met were detected by Real Time Fluorescence Quantitative PCR(FQ RT-PCR) method in 56 cervical carcinoma tissues,30 cervical intraepithelial neoplasmâ…¢(CINâ…¢)tissues and 27 normal cervical tissues respectively. (2)Levels of serum HGF and VEGF-C were determined by enzyme linked immunosorbent assay (ELISA) in 15 cases with normal cervix and 73 cases cervical cancer. (3)To analyze difference expression of VEGF-C,HGF and c-Met between the groups and difference in clinical stages and the clinical pathologic characters (clinic pathology,differentiation,tumor size,depth of stromas invasion,lymph node metastases). To analyze the correlation of VEGF-C and HGF ,HGF and c-Met.Results (1) The positive expression of HGF mRNA,c-Met mRNA and VEGF-C mRNA increased remarkably from normal tissue to CINâ…¢and then to cervical carcinoma (P<0.05). (2) The concentration of serum HGF in cervical cancer was significantly higher than normal cervix. (3) Levels of VEGF-C mRNA,HGF mRNA and c-Met mRNA ,and those of serum HGF and VEGF-C show increasing with the development of clinic stage(P<0.05). (4) Levels of mRNA expression and concentration of serum of VEGF-C and HGF were correlated with grade depth of stromas invasion and lymph node involvement ( P<0.05). Higher Levels of serum c-Met in cervical cancer was correlated with tumor size,depth of stromas invasion and lymph node involvement ( P<0.05). (5) Expression of HGF mRNA was correlated with Squamous cancer, VEGF-C mRNA was highly expressed in poorly differentiated cervical cancer. (6) HGF mRNA and c-Met mRNA expression was positively correlated(r=0.476,P<0.01). Expression of VEGF-C and HGF in the serum and tissues were positively correlated(r=0.460 and 0.303,P<0.01).Conclusion (1)Over expression of HGF and VEGF-C, to investigate the expression level in the serum and Cervical biopsy specimens would provide guidance to preoperative diagnosis and prognosis. (2)In cervical carcinoma, the expression of these three genes were positive correlated with the clinical stage,tumor size,the invasion of deep muscular layer and the lymph node metastasis(P<0.05). These three genes may together participate in the invasion and metastasis of cervical carcinoma. (3)The expression and their consistency of HGF and c-Met could evidence further that HGF may be play a role in malignant tumors through a combination of receptor c-Met. (4) Serum HGF and VEGF-C expression was positively correlated.To investigate HGF and VEGF-C together would be facilitated to the treatment and prognosis of cancer. |
