Objective To study the protective effect of concha haliotidis extractive on the oxidativedamage human lens epithelial cells cultured in vitro.Methods Cultured human lens epithelial cells in vitro were divided into control group, thepositive control group(hydrogen peroxide group) and the treated group.Oxidative damagemodel was established by culturing human lens epithelial cells with hydrogen peroxide. Thehuman lens epithelial cells in the blank control group were cultured with.The human lensepithelial cells in the positive control group were cultured with hydrogen peroxide.The humanlens epithelial cells in the treated group were cultured with hydrogen peroxide and differentconcentrations of concha haliotidis extractive. On the first,the third and the fifth day, theproliferation of cultured human lens epithelial cells were detected with CCK-8, cellularproliferation and the morphological changes were observed with interted phase contrastmicroscope.On the third day the level of homogenates-superoxide dismutase(SOD),glutathione(GSH) and malondialdehyde(MDA)were detected with chemical colorimetric,theapoptosis rate was deceted with flow cytometer.Results①The proliferation of HLECs varied at different time points.The different amonggroups was statistical significance(1d, F=23922.421,P<0.05;3d,F=120605.864, P<0.05;5d,F=150939.452,P<0.05). H2O2reduced the proliferation of HLECs, while concha haliotidisextractive promotedd its vitality,at certain range of time and concentrations with a manner oftime dependent manner and concentration dependent.The best manner was0.1%at the thirdday.②A fter addingH2O2, the SOD and GSH level of HLECs reduced but the MDA increased.Concha haliotidis decrea-sed the antioxidant and increased the lipid peroxidation in oxidativedamaged HLECs. The difference between had a statistical significance(SOD: F=983.042,P<0.05; GSH: F=444.446, P<0.05; MDA: F=830.523, P<0.05).③Comparing to the positivecontrol group, the treated group had a lower apoptosis rate. The difference had a statistical significance (F=5139.030,P<0.05, P<0.05).④The chromatin of the positive control groupconcentrated and aggregated obviously,while that of treated group lightened.Conclusion The concha haliotidis own the protective effect for the oxidative damage humanlens epithelial cells cultured in vitro which increases intracellular antioxidant levels,reduces thegeneration of hazardous products and promotes proliferation.
