The Relations Between Heparanase With Invasion And Metastasis Of Human Beast Tumor And The Effects Of Ulinastatin | | Posted on:2013-01-02 | Degree:Master | Type:Thesis | | Country:China | Candidate:N Wang | Full Text:PDF | | GTID:2234330374978063 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Objective:To study the relations between Protein Heparanase(Hpse)ã€Hyaluronidase-1(HYAL1)and Cellular adhesive molecular CD44v6andinvasion and metastasis of human breast cancer cell MCF-7(the estrinreceptor is positive) and MDA-MB-231(the estrin receptor is negative)andthe effect of ulinastatin. Methods:The breast cancer cell line MCF-7and MDA-MB-231cultured in vitro were treated in randomly divided:1.Control,2.UTI at thelow-dosage group,3.medium-dosage group,4.high-dosage group,5.moderate dosage of UTI with Taxotere(TXT).the proliferation ability ofcells are tested by MTT assay. Western blotting applications are used tomeasure the expression of Hpseã€HYAL1and CD44V6;The invasion abilityof the cell lines are tested by Transwell chamber assay with Matrigel;Themigration of the two cell lines are examined by Transwell chamber. Results:The expression of Hpse〠HYAL1and CD44V6wassignificantly decreased compared with the control group, and the effect ofthat received two medicament simultaneously is superior to that UTI alone(P﹤0.05); compared with the control cells,UTI inhibit the invasionand migration of the breast cancer cell MCF-7and MDA-MB-231,moresignificant joint UTI and TXT (P﹤0.05).Conclusions:UTI can inhibit the invasion and metastasis of the breastcancer cell, the mechanism may be associated with UTI reduced theexpression of Hpseã€HYAL1and CD44V6. | | Keywords/Search Tags: | Breast cancer, Heparanase, Hyaluronidase-1, CD44V6, Ulinastatin | PDF Full Text Request | Related items |
