An Investigation Of Serum Sex Hormones And Ovarian Morphology On Polycystic Ovary Syndrome Type Rats Treated With Promoting Kidney Clearing Liver And Blood Circulation Decoction

Objective:Through observing the change of serum sex hormones levels, vaginal exfoliated cells and ovarian morphology on polycystic ovary syndrome type rats induced by letrozole. to discuss the intervention effect and mechanism of Tonifying Kidney Clearing Live and Blood Circulation Decoction treating PCOS.Methods:Sixty six weeks old clean level female SD rats weighted170-200grams were selected. Rats were randomly divided into normal control group and modeling group. The PCOS rat model was made using letrozole. After the models were made, rats of modeling group were randomly divided into model control group, CC group, medium dose of Nourishing Kidney Clearing Live and Blood Circulation Decoction (NCCD) and high dose of NCCD group. For each group including12rats, medicine was administered intragastrically. Normal group was conventional breeding without irrigation stomach stimulation. Water was given2ml respectively in model group, once a day for20days. Rat in CC group was given CC suspension (concentration:0.75mg/ml)2ml respectively once a day for5days, after taking daily water irrigation for15days. The rats of medium, high dose group were offered solution2ml respectively, once a day for20days. The medium, high dose solution contains NCCD1.5g/ml,3g/ml severally. Ovarian pathology and serum sex hormones were tested for each rat after20days.Results:After medicine affecting, the weight of rat dropped significantly for treatment group compared to model group (P<0.01). But modeling rats presented the state of growth. Compared with normal group, the serum LH. T and FSH level of model rats increased dramatically (P<0.01), as well as serum E2and P level fallen clearly(P<0.01). Compared to model control group, the serum LH. T and FSH level of rats in treatment group decreased observably (P<0.01), while serum E2and P level dropped obviously (P<0.05).The level of serum LH. T and FSH was on the high side for CC group compared to NCCD group (P<0.01). when serum E2level was slightly low (P<0.05). Compared with normal group, the ovarian weight of model group increased signally. But rats ovarian weight in treatment group clearly fallen comparing with model group (P<0.05). Compared to normal group, the uterine weight of modeling rats reduced, but no significantly difference (P>0.05). Compared with model group, there were more follicles at all levels in the ovary of NDDC group, as well as advantage follicles were more obvious, granule cell layer were incrassated. the amount of corpus luteum was increased, and cystiform follicles decreased or disappeared.Conclusion:The PCOS rat model was made using letrozole, was greatly similar to the clinical characteristics of human PCOS not only in the change of endocrine but also in ovarian morphological change. It means this way is a kind of ideal modeling methods. NCCD can improve the hyperandrogenism of PCOS model rats, significantly reduce serum LH and T levels, as well as increase E2and P contents, and that regulate the function of reproductive axis, to promote the normal development and maturity. It also can improve the microcirculation of ovarian local to make ovarian coated thin in favor of ovulation of follicles, aiming to fundamentally improving the polycystic condition of ovary. The effect of high dose of NDDC group is best of all.
