Background&Purpose: Thymoma is one of the most commontumors of the anterior mediastinum and it is from thymic epithelial cells,because of their microscopic structure of complex and volatile, there is aninconsistency between the pathological and prognosis, and it has thecharacteristics of local external intrusion spread, recurrence and distantmetastasis,so their performance in the biological behavior of low-grade,thus, the clinical thymoma is a low-grade malignant tumor or malignanttendency. U.S. National Cancer Institute (NCI) survey showed that theincidence rate of thymoma population was0.15/10000,there was nosignificant difference between the gender and the highest incidence of40to50-year-old crowd. The pathogenesis of thymoma is not very clear now,and the clinical histopathological examination can not fully reveal thebiological behavior of thymoma. In recent years, with the application ofmolecular biology techniques and a variety of tumor molecular markershave been found to provide a more comprehensive and intentionalbiological information for the probe into the biological behavior of tumordevelopment, thereby assisting the pathological diagnosis of thymoma,and have achieved some break through. We applicatedimmunohistochemistry(SP method) to detect the expression of PTEN、Ki-67and EGFR in thymoma tissue to explore whether the three and theWHO histological type, Masaoka stage has associated with MG relationship,from the clinical pathology point of view and by therelationship among the three and its expression in thymoma to reveal itsrole in the pathogenesis of thymoma and possible mechanisms to provideobjective judgment indicators for clinical diagnosis and treatment.Methods: In this experiment, we collected52cases of thymoma and5cases of normal thymic tissue as an object of study, all tissue samplesderived from the First Hospital of Jilin University, Bethune. Byimmunohistochemistry (SP) to detect the expression of PTEN、Ki-67andEGFR in the different installments the thymoma tissue and normal thymictissue,and the results with SPSS17.0software for statistical analysis, χ2test was used for significance test and the correlation analysis usingspearman rank correlation coefficient, P=0.05level of inspection.Results: In the52thymoma tissue samples of the detected, PTEN、Ki-67and EGFR positive expression rates were as follows:71.15%,53.85%,48.08; the expression rate of three in the five cases of normalthymic tissue were20.00%,0.00%, and100%respectively,the expressiondifferences of three were statistically significant (P <0.05), there is aclose relationship between the expression level of EGFR and Masaokastaging,With the the installments progress positive expression rategradually increased, and the difference was statistically significant (P<0.05), there is no correlation with WHO pathological and histologicaltype (P>0.05),there is a close relationship with Ki-67positive expressionlevels and the WHO histological type and the Masaoka staging,With theprogress of staging the positive expression rate gradually improve, thereis a statistically significant difference (P <0.05); the positive expressionof PTEN decreased gradually with the progress of the the WHOhistological type and Masaoka staging, that there is a statistically significant difference (P <0.05). Ki-67&EGFR expression in thymomawas positively correlated (P <0.05); Ki-67&PTEN expression inthymoma negatively correlated (P <0.05). the positive expression rates ofPTEN, Ki-67and EGFR with the thymoma patients whether to mergeMG has no correlation, and that the difference was not statisticallysignificant (P>0.05).Conclusion: There are multiple genetic variants in the developmentof molecular mechanisms of the thymoma, it played a role in whichLoss of expression of PTEN and overexpression of EGFR, Ki-67,theexpression of PTEN, EGFR and Ki-67in thymoma may reflect the WHOhistological type of thymoma and Masaoka staging,it can be as aprediction of tumor invasion of indicators for the early diagnosis ofthymoma and pathological staging help.It was unrelated. with the proteinexpression of PTEN、Ki-67and EGFR with whether to merge MGthymoma patients. PTEN、Ki-67and EGFR act synergistically in thedevelopment of thymoma, The combined detection of three canconducive to a better assessment of thymoma biological properties whileprovide objective indicators for the diagnosis and prognosis of thymoma.
