Coptis chinensis, which belongs to Ranunculaceae, is the dried rhizome of perennial herb Coptis chinensis Franch. or Coptis. deltoidea C.Y. Cheng et Hsiao or Coptis teeta wall. It is important tranditional Chinese medicine which used in clinical commonly. The extraction of active ingredients and removal of harmful substances from traditional Chinese medicine are important parts in the production process of pharmaceutical, which also are perfect method to ensure the quality of traditional Chinese medicine.In this research, we use the yield rate of berberine in coptis pieces as the target to establish the optimum extraction technology using organic solvents refluxing. To study on heavy metal removal from Coptis chinensis Franch., atomic absorption spectrophotometry was employed to determine the content of cadmium and lead. At the same time, the atomic fluorescence spectrophotometry was used to determine the content of mercury and arsenic.1. In the study on the extraction technology of berberine in Coptis chinensis Franch. using organic solvents refluxing, based on the results of the single factor experiments, the orthogonal test and response surface method were employed to systematically study the three important effects which impact on the absorbance of extraction greatly. According to the results of the test, the absorbance of berberine×1000as response value to set up response surface in order to establish the optimum extraction technology of berberine in Coptis chinensis Franch were53%ethanol concentration,62℃extraction temperature,1:11solid/solvent ratio and extract for1h×2. Under the optimal extraction condition, berberine yield was6.12%.2. Study on cadmium removal from Coptis chinensis Franch., atomic absorption spectrophotometry was employed to determine cadmium content, mixed acid digestion was used to pre-treatment sample. Results showed that the best treatment effect is3%ammonia for six hours+3%NH4NO3for six hours, the content of cadmium is0.0376(mg/kg)<0.3(mg/kg), the content of berberine is5.2%.3. Study on lead removal from Coptis chinensis Franch., atomic absorption spectrophotometry was employed to determine lead content, mixed acid digestion was used to pre-treatment sample. Results showed that the best treatment effect is5%acetic acid for six hours+0.3%HNO3for six hours, the content of lead is0.5627(mg/kg)<5.0(mg/kg), the content of berberine is4.8%.4. Study on mercury removal from Coptis chinensis Franch., atomic fluorescence spectrophotometry was employed to detemine mercury content mixed acid digestion was used to pre-treatment sample. Results showed that the best treatment effect is3%ammonia for six hours+0.01%acetone for six hours, the content of mercury is0.0452(mg/kg)<0.2(mg/kg), the content of berberine is5.0%.5. Study on arsenic removal from Coptis chinensis Franch., atomic fluorescence spectrophotometry was employed to detemine arsenic content, mixed acid digestion was used to pre-treatment sample. Results showed that the best treatment effect is0.3%NaOH for six hours+3%Na2CO3for six hours, the content of arsenic is0.2158(mg/kg)<2.0(mg/kg), the content of berberine is4.8%.
