| Objective:To explore the expression of heat Hest Shock Protein90β(HSP90β) in gastric adenocarcinoma, gastritis and adjacent tissues, then analysis of the correlation between HSP90βand tumor cell proliferation and clinicopathological parameters, in the end we discuss its relationship with tumor cell invasion and metastasis and prognosis.Methods:Collect archive wax blocks to made paraffin tissue microarrays and use immunohistochemical PV-9000two-step method to detect the expression of HSP90β, cell proliferation factor Ki-67, matrix metalloproteinase MMP-2and inhibitor of TIMP-2and adhesionmolecule CD44v6in60cases of gastric adenocarcinoma and20cases of gastritis. Collect11groups of gastric adenocarcinoma and adjacent tissues, the expression levels of HSP90β mRNA were detected with RT-PCR method. SPSS19.0software is used to analyze the expression of HSP90βin gastric adenocarcinoma, the correlations between HSP90βand other indicators, the relationship between them and the clinicopathological parameters, survival analysis and prognosis of60patients with gastric adenocarcinoma.Results:1.The expression of HSP90βin gastric adenocarcinoma is higher than that in gastritis tissues (P<0.01), and the positive rate in poorly differentiated gastric adenocarcinoma is much higher than that in well-differentiated and moderately differentiated gastric adenocarcinoma.The difference was statistically significant (P<0.05). While it is independent of the parameters of age, gender, tumor invasion and lymph node metastasis (P>0.05).2. The expression of HSP90β in gastric adenocarcinoma with Ki-67and CD44v6, there is a positive correlation (P<0.05), with no significant correlation between HSP90β with the expression of MMP-9and TIMP-2(P>0.05).3.The survival time of patients of gastric adenocarcinoma with positive expression of HSP90β is shorter than those with negative expression (P<0.05).4. The expression of HSP90β mRNA amount in gastric adenocarcinoma is higher than the expression level in the adjacent tissues (P<0.05).Conclusions:1.HSP90P is significantly up-regulated in gastric adenocarcinoma and its expression is positively correlated to cell proliferation status.2.The worse histology with the higher positive expression rate of HSP90βin gastric adenocarcinoma, the high expression of HSP90β indicates a poor prognosis.3.The high expression of HSP90β may indicate the strong ability of the invasion and metastasis of gastric adenocarcinoma. |
