Objective:Observe electroacupuncture Heart Meridian of Hand-Shaoyin acupuncture point focal in focal cerebral ischemia rats, to study the effects of nerve deficit symptoms, cerebral infarction volume and infarction rate, serum and brain levels of TGF-(3, to explore the nervus protection function and mechanism about ischemic brain injury with electroacupuncture Heart Meridian of Hand-Shaoyin, and to provided some experiment basis about electroacupuncture Heart Meridian of Hand-Shaoyin treat Cerebrovascular disease and the relationship among Heart Meridian of Hand-Shaoyin and brain.Methods:50male rats randomly divided into5groups; The test factors is electroacupuncture Shenmen and Shaohai meridians of Heart Meridian, compared with control group, model group、 sham operation group and lung group. The neurological deficit score were gone at6h、24h、48h and72h, after cerebral ischemia for each rat; Draw materials after all treatment over, detected cerebral infarction volume by TTC staining; Serum concentrations of TGF-β by enzyme-linked immunosorbent assay; TGF-β expression in brain tissue by Sreptavidin-biotin complex(SABC); Brain tissue of TGF-β by Quantive-PCR(Q-PCR).Result:1. The neurological deficit score of the Heart Meridian group has lower than the model group at72h(P<0.01). This score of the Heart Meridian group at72h has lower than6h(P<0.05), but completely opposite to the model group.2. The cerebral infarction volume and infarction rate of Heart Meridian group has observably less than the model group(P<0.01), as the same of Lung Meridian group on the cerebral infarction volume (P<0.05), but no diversity on cerebral infarction rate(P>0.05).3. The serum and brain levels of TGF-β of model group, Heart Meridian group and Lung Meridian group are higher than control group and sham operation group(P<0.01), and the Heart Meridian group is higher than the Lung Meridian group(P<0.01).4. Compared with the control group, the expression of TGF-β mRNA in the cerebral tissue has higher than the model group(P<0.05), and obviously higher on the Lung Meridian group and the Heart Meridian group(P<0.01).This expression on the Heart Meridian group has obviously higher than the model group(P<0.01),and the Lung Meridian group has higher than the model group, too(P<0.05).Conclusion:1. Three days after cerebral ischemia injury, the cerebral lesion is still in the aggravating. This injury can stimulate the body ourself release TGF-β in serum and brain tissue.2. Electroacupuncture Heart Meridian of Hand-Shaoyin can reduce the neurological deficit score and cerebral infarction volume and infarction rate, This function which protected and repaired the neuron may related to electroacupuncture Heart Meridian can promote TGF-β release in serum and brain tissue.3. Electroacupuncture Heart Meridian can improve the MCAO rats neurological deficit score, minus the cerebral infarction volume, which more than Lung Meridian. The mechanism is the specificities of acupoints or not, worthy of futher investigation.
