Objective:TO study the expression of Notch1in ovarian cancer tissue and its clinicalpathology meaning. TO study the influence of dihydroartemisin(DHA) on theproliferation of human ovarian cancer cell line Skov3and the express of Notch1proteins.Methods:The expressions of Notch1were detected in12cases of normal ovarianepithelial tissue,21cases of ovarian cystadenoma and63cases of ovarian epithelialcarcinoma by ElivisionTMplus two-step of immunohistochemical staining.Skov3wascμltured in different concentrations of DHA(0,5,10,20,40,80μmol/L) in vitro. MTTassay was used to measure the effects of different concentrations ofDHA on ovariancancer cells line Skov3. RT-PCR and Western Blot assay were used to measure thedifferent levels of Notch1protein expression and Notch1mRNA in Skov3cells whichwere processed by different concentrations of DHA after24h.Resμlt:1.The positive expression rates of Notch1in normal ovarian epithelial tissue,ovarian cystadenoma and ovarian epithelial carcinoma were16.7%,38.1%and73.0%respectively.Statistical differences among the three groups were significant(P<0.05).The expression of Notch1protein was unrelated to the patient’s age andhistological type (P>0.05), but was related to pathological grade,stage of the tumorand lymph nodes metastasis(P<0.05).2.Cell line skov3of human ovarian cancer was inhibited markedly by differentconcentrations of DHA (0,5,10,20,40,80μmol/L) after processing at different times(24h,48h,72h). The proliferation of skov3was significantly suppressed in dose-andtime-dependent manners. The comparison between the groups and the comparisonbetween each group and control group were significant differences (P<0.05).3.Cell line skov3of human ovarian cancer was inhibited markedly by differentconcentrations of DHA (0,5,10,20,40,80μmol/L) after processing24h.With the increasing of DHA concentration, the expression of Notch1mRNA in human ovariancancer cell was down regμlated.4.Cell line skov3of human ovarian cancer was inhibited markedly by differentconcentrations of DHA (0,5,10,20,40,80μmol/L) after processing24h.With theincreasing of DHA concentration, the expression of North1protein in human ovariancancer cell was down regμlated.Conclusions:1.Notch1plays an important role in occurrence and development of ovarianepithelial cancer.2.DHA may suppress the proliferation of human ovarian cancer cells Skov3.3.DHA may down-regμlated the expression of Notch1in Skov3cells.
