Ovarian cancer is one of the common gynecologic malignancies because the vast majority patient is already advanced when first diagnosed, the mortality rate is the highest in the domain. At present, and patients after ovarian cancer surgery or who can not be operated are required to have a chemotherapy to improve quality of life,increase the survival rate, reasonably choosing the chemotherapy drugs and its compatibility programme is particularly important. These years, cisplatin is the first-line chemotherapy drugs for ovarian cancer, because its side effect and resistance is obvious due to the effect of its chemotherapy is poor, to search for the new combinated methods,particularly searching for more effective,lesser side effects,cheapper drug have been a priority in research. Firstly, this paper reveals the regularity of its expression in benign, borderline and malignant ovarian epithelial tumors by measuring vascular endothelial growth factor (VEGF) in epithelial ovarian tumor tissues,and further determines expression of VEGF in different ovarian cancer tissue , including different pathological types,different pathological grades, clinical stages,different ascites quantity, different lymph node metastasis, attempting to clear the relationship in the expression of VEGF and epithelial ovarian cancer clinical phases,the degree of progress. and explores whether depressing the expression of VEGF is one of the mechanisms of role of MLT resisting human ovarian cancer SKOV3 cells proliferation and pro-apoptotic ,and further studies vitro effect of MLT,DDP alone and combined application in inhibiting human ovarian cancer SKOV3 cells .The purpose of this study include the following two aspects:1. Immunohistochemical staining technique have a result of expression in vascular endothelial growth factor (VEGF) of benign, borderline and malignant ovarian tumor clear the relationship in the expression of VEGF and epithelial ovarian tumors clinical phases,the degree of progress and so on.2. Using microscopy observational method,MTT assay to kown how the ovarian cancer SKOV3 cells in vitro growth inhibition by MLT,DDP and the two drugs combinated are,and applying immunocytochemical methods to measure the expression of VEGF in the ovarian cancer SKOV3 cells in vitro when before given these drugs or behide .Exploreing above kinds of results make an analysis judgement whether decreasing expression of VEGF in ovarian cancer SKOV3 cells in vitro is one of the mechanisms of inhibiting cancer cell growthThrough research we have the following results:1. The expression of VEGF gradually increases in the benign, borderline and malignant epithelial ovarian tumors .2.In Ovarian cancer, the patients whose condition is in late stage,a large amount of ascites and the existence of lymph node metastasis , the expression of VEGF are higher than those in a stage early,less ascites,lymph node metastasis , but in different pathological types there isn,t obvious expression difference.3. Morphological observations: SKOV3 cultured ovarian cancer cells in the inverted microscope, the applying drug group than the unapplying drug group :Cells, adherent capacity is significantly weakened, and cell shape changes small and round, refraction is weakened significantly . This situation is more obvious with the increase of drug concentration,the two drug jointed . Therefore, the judge from perceptual knowledge is that both MLT and DDP are able to inhibit ovarian cancer SKOV3 cell growth and proliferation, furthermore the two drugs jointed can enhance this effects. Malignant tumor cell growth,proliferation was inhibited significantly,and its apoptosis was increased by MLT and combined with DDP . Determination of VEGF has important reference value the clinical diagnosis and prognosis.4. Immunocytochemical staining:expression of VEGF are all descreasd in ovarian cancer SKOV3 cells by MLT,DDP single-drug group and their combination group , furthermore, the combination of the two drugs has synergistic action on the down-regulation expression of VEGF .5. MTT assay detect: according to inhibition rate formula, the two drug interaction index formula, the results showed that 0.5 mmol / L, 1.0mmol / L and 2.5 mmol / L MLT and 25 mg / L, 50mg / L DDP Joint Application have synergies, and the inhibitory rate increased with increasing concentration.Conclusion :1. The expression of VEGF gradual increases in benign, borderline and malignant epithelial ovarian tumors.2. The expression of VEGF is related with the period of ovarian cancer, ascites amount , lymph node metastasis condition, but it has nothing to do with the pathological type. Determination of VEGF has important reference value the clinical diagnosis and prognosis.3. MLT plays inhibition action on ovarian cancer cell growth, proliferation ,the down-regulation in the expression of VEGF may be one of the mechanism.4. The combination of MLT and DDP has synergistic action on the down-regulation expression of VEGF in Ovarian cancer skov3 cell. DDP is be compatibilited with MLT can reduce achieve the same effect with Much more DDP ,so we can play Appropriate amount MLT to reduce the dosage of the DDP , thus we achieve our goal Of reducing DDP,s side effect.
