Objective:To investigate the mechanism of action of Dihydroartemisinin (DHA) oninducing apoptosis of the human ovarian cancer cell SKOV3in vitro,and study theDihydroartemisinin enhance the chemotherapy sensitivity of the human ovariancancer cell SKOV3to Carboplatin.Methods:⑴Human ovarian cancer SKOV3cells were cultured in vitro,the cytotoxic effecton the proliferation was determined by MTT colorimetric method.⑵The effect of apoptosis was detected by flow cytometry assay.⑶Human ovarian cancer SKOV3cells were intervened in DHA,Bcl-2、BaxmRNA expression were analyzed by RT-PCR assay.Results:⑴DHA Significantly inhibited the proliferation of SKOV3cells,induced theirapotosis in a time-concentration dependent manner,each concentration group wasstatistically different compared with the control group(P<0.05).After SKOV3cellstreated with Dihydroartemisinin,the different IC50was117、5.677and23.382μmol/L(24h、48h、72h).⑵Dihydroartemisinin could inhibite the growth and induce apoptosis of theovarian cancer SKOV3cells,DHA exhibits a dose and time-dependment cytotoxicity(P<0.05);After SKOV3cells treated with Carboplatin,the IC50was189mg/L(24h);After SKOV3cells treated with Carboplatin combined with low doses ofdihydroartemisinin,the IC50dropped down to61.289mg/L(24h),the sensitizationmultiples was3.08.⑶Dihydroartemisinin could reduce the expressions of Bcl-2and increase heexpressions of Bax.The absorbance value of each concentration compared with the control group,was statistically different(P<0.05).Conclusion:⑴Dihydroartemisinin could induce Apoptosis in Human Cancer SKOV3Cellsin vitro.⑵Dihydroartemisinin could enhance the chemotherapy sensitivity toCarboplatin.⑶Dihydroartemisinin could induce the apoptosis in SKOV3cells by up-regulating Bax protein and down-regulating Bcl-2gene.
