Purpose:Through exploring the influence of the traditional Chinese medicine Pen Yan Fang on the expression of Hemarheology and GM-CSF about the SPID rat models to provide the experimental basis for the clinical application.Method:Choosing124SD female rats as the subjects and randomly extracting10female rats as the normal group and10female rats as the sham-operated group, later using the mixed bacteria inoculation method to establish SPID rat models; After successfully making models, rat models will be randomly divided into model group for15rats, therapeutic group for15rats, Pen Yan Fang high, medium and low dose group with each of10rats. Each group are given corresponding medicines for21days then will be the blood sampling step. Observing the change of the uterus organization morphology under HE dyeing. The hemarheology indexes were measured by blood viscosity receptor, then using the ELISA method to observe the GM-CSF expression of all groups of rats.Results:1. The uterus organization morphology of rat models was observed under HE dyeing, which is consistent with chronic inflammation’s performance. After the treatment, the condition of all groups had various degrees of improvement, and Pen Yan Fang high-dosage group had the most significant improvement (P<0.05). 2. Hemarheolog:All the values of whole blood viscosity has no obvious expression in normal group and sham-operated group, but the model group exists high expression, with comparative differences which has remarkable statistical significance among the three groups (P<0.01). In the high-dosage and medium-dosage group, the whole blood viscosity were remarkable reduced compared with the model group of rats (P<0.05or P<0.01). The therapeutic group whole blood viscosity after21days is obviously below the low-dosage group(P<0.05or P<0.01).The low-dosage group’s high shear viscosity and low shear viscosity is obviously below the model group (P<0.05), but it has no statistical difference comparing with the medium shear viscosity (P>0.05); Under the same dosage, the whole blood viscosity (medium and low shearing) in medium-dosage group were lower than that in co therapeutic group (P <0.05or P<0.01),but has no statistical difference in the high shearing(P>0.05).3.GM-CSF has no obvious difference in normal group and sham-operated group, but the model group exists high level, with comparative differences which has remarkable statistical significance among the three groups (P<0.01); The level of Pen Yan Fang high-dosage group is obviously below the model group(P<0.05or P<0.01), while also has meaningful comparative difference with the therapeutic group(P <0.05); The level of Pen Yan Fang low-dosage group was significant lower than model group, but has no statistical meaning comparing with the therapeutic group (P>0.05).Conclusions: 1. SPID rat models can be successfully copied by using mixed bacteria inoculation method.2. Pan Yan Fang can improve the structure and function of the uterus of SPID model rats.3. Pen Yan Fang can change the whole blood viscosity, and can reduce the level of the GM-CSF, thus plays its anti-inflammatory effect, which might be one of the mechanism of treating SPID.
