The Cang Ai volatile oil, Compound preparation, summarizes tutor’syears of experience in clinical medicine, the previous studies have shownthat the volatile oil have a great significant inhibitory effect on the AURIcommon strains such as staphylococcus aureus, Group A streptococcuspneumoniae and Group B Streptococcus. The nasal inhalationadministration can effectively improve the SIgA content in saliva of mice,protect the upper respiratory tract from smoke damage, prevent upperrespiratory tract infection. On the basis of the theory and preliminarywork, the subject carries our deep experimental researches in Cang Aivolatile oil of Lung Qi Deficiency’ Syndrome on the Regulation ofImmune in Mice.Objective:Explore the immune regulation function and the mechanism ofaction with The Cangai volatile oil of lung qi deficiency rats, in order toprovide a novel and effective treatment of lung qi deficiency syndromefor the clinical treatment.Method:1. By collecting the model building methods of lung qi deficiencysyndrome animal in the past, Choose two lung qi deficiency syndromeanimal model methods in common use to copy. Compare each model’sdiagnostic indicators of objective. Pick up the one which have high sensitivity and with good reproducibility.2. Reference the median lethal dose of Cangai volatile oil tocalculate the Administration dosage for the low dosage, middle dosageand high dosage. Model building and give drugs at the same time, alsocompare the mode of administration between gastric perfusion and nasaladministration. Model building for31days, take out blood serum, lungand pharyngeal of rats, ELISA determination of rat blood serum IgE、TNF-α、IFN-γ、IL-1β levels and use the flow cytometry technology todetect the expressions of T lymphocyte subtype. Observe the pathologicalsection of rats’ lung and pharyngeal.Result:1.According to a lung qi deficiency syndrome diagnostic criteria of‘Guidelines for clinical research of traditional Chinese medicine’ in2002,the simple smoke group and LPS+smoked compared with normal group,the two model groups’ signs and symptoms of lung qi deficiency is veryobviou(sP<0.01), the latter group is wors(eP<0.01). And the two modelgroups’ pathological changes in the lung and pharyngeal are moreobvious. Positive lymphocytes could also be seen in the lung andpharyngeal of the two model groups. The LPS+smoked group alsoappeared the pathological changes of the alveolar septa in width,pulmonary bullae formation and small blood vessels.2. The CD4+lymphocyte proportion of Model groups is decline sharply compared with the normal group and showed statisticallysignificant(P<0.05). The two modes of administration gastric perfusionand nasal administration could efficiently regulate the T cell subsets’proportion of rats’ whole blood who is the lung qi deficiency syndrome init, itcan make the T lymphocyte subsets ratio close to normal.3. The IFN-γ content of Model groups are decline sharply comparedwith the normal group and showed statistically significant(P<0.05,P<0.01).The administration gastric perfusion and nasal administrationgroups of middle dosage and high dosage with the dose of Cangai volatileoil were significantly higher than the two model groups, showedstatistically significant(P<0.05).4. The IL-1β content of Model groups are significantly highercompared with the normal group and showed statistically significant(P<0.01,P<0.05). The administration gastric perfusion groups of low dosage,middle dosage and high dosage with the dose of Cangai volatile oil weredecline sharply than the model groups, showed statistically significant(P<0.05). The nasal administration groups of middle dosage and highdosage with the dose of Cangai volatile oil were decline sharply than themodel groups, showed statistically significant (P<0.05). Theadministration gastric perfusion and nasal administration groups ofmiddle dosage and high dosage with the dose of Cangai volatile oil weresignificantly higher than the two model groups, showed statistically significant(P<0.05).5. The TNF-α content of Model groups compared with the normalgroup showed no significant difference(P>0.05).The administrationgastric perfusion and nasal administration groups of low dosage, middledosage and high dosage with the dose of Cangai volatile oil comparedwith the two model groups showed no significant difference(P>0.05).6.The IgE content of Model groups compared with the normal groupshowed no significant difference(P>0.05).The administration gastricperfusion and nasal administration groups of low dosage, middle dosageand high dosage with the dose of Cangai volatile oil compared with thetwo model groups showed no significant difference(P>0.05).7. Through the observation of pathological, The administration gastricperfusion of low dosage, high dosage and the nasal administration groupsof low dosage, high dosage with the dose of Cangai volatile oil canreduce the pharyngeal inflammation cells, and also can reduce congestionand edema position. The administration gastric perfusion and nasaladministration groups of high dosage with the dose of Cangai volatile oilcan reduce the infiltration of inflammatory cells in the alveolar wall,Improve the congestive position of lung tissue and repair damagedmucosa organizations.Conclusions:1. The signs and symptoms of lung qi deficiency and the pathological changes is very obvious compared with the normal group, the model copysucceed. And the LPS+smoked group worse than the simple group, it’sappeared the pathological changes compared with the simple group, theLPS+smoked group more in line with the diagnostic index of lung qideficiency syndrome group, so use it as a study pulmonary qi deficiencysyndrome animal model building methods.2. Prevention of Cangai volatile oil, can effectively improve the T cellsubgroup CD4+/CD8+ratio of the rats who is the lung qi deficiencysyndrome in it, and it can promote cell immune adjustment.3. Prevention of Cangai volatile oil, can effectively improve the IFN-γcontent of rats’ whole blood serum who is the lung qi deficiencysyndrome in it, and strengthen the body’s immune regulating function.4. Prevention of Cangai volatile oil, can reduce the content of I L-1βof rats’ whole blood serum who is the lung qi deficiency syndrome in it,and effectively inhibited the inflammatory response of the lung qideficiency model rats.5. Prevention of Cangai volatile oil, can reduce the infiltration ofinflammatory cells in the alveolar wall, Improve the congestive positionof lung tissue and repair damaged mucosa organizations.6. The two modes of administration gastric perfusion and nasaladministration with the Cangai volatile oil all have the same effect, whichone is better need a deeply study in the further.
