The Effects Of Yiqi Huayu Qingre Party And Its Separate On Rat MsC Apoptosis, Expression Of Ubiquitinated Proteins

Objective: By observing the effects of Yiqi Huayu Qingre party and its separate onRat mesangial cells apoptosis and apoptosis related genes bax,bcl-2,as well as theubiquitinated protein in vitro circumstance stimulated by lipopolysaccharide(LPS)，probe the mechanism of Yiqi Huayu Qingre party in the treatment of mesangialproliferative glomerulonephritis(MsPGN),and compare the intensity of action ofdifferent kinds of traditional Chinese medicine.Methods: The rat MsC stimulated by LPS in vitro circumstance to make theproliferative model was divided randomly into eight equal groups: normalgroup;model group;control group; contained-yiqi serum group; contained-huayuserum group,contained-qingre serum group;contained-compound serumgroup;contained-Tripterygium wilfordi serum group,repectively joining in all kinds ofmedicated serum continue to develop,using Western-blot method to detect theeffects of the medicaed serum on the expression of apoptosis related genesbax,bcl-2,as well as the ubiquitinated protein; using TUNEL method to detect theeffects of all kinds of medicated serum on MsC apoptosis.Results: Yiqi Huayu Qingre party and its separate can induce the apoptosis of ratMsC in vitro circumstance by improving the ratio of Bax/Bcl-2, thecontained-compound serum group and the contained-Tripterygium wilfordi serumgroup were stronger,the effect of the contained-huayu serum group and thecontained-qingre serum group was better than the contained-yiqi serum group (P <0.05); Yiqi Huayu Qingre party and its separate can reduce the expression ofubiquitinated protein in MsC of MsPGN, the contained-compound serum group wasthe strongest, the effect of the contained-huayu serum group and the contained-qingreserum group was better than the contained-yiqi serum group(P <0.05), betweenwhich was no significant difference (P＞0.05).Conclusion: Yiqi Huayu Qingre party and its separate each can induce the apoptosisof MsC and reduce the expression of ubiquitinated protein,the effect of huayu and qingre drugs is better than that of yiqi medicines.
