| Objective:Through observed the clinical effects of curing pediatric hormome sensitive nephrotic synrome with Yiqi Huayu Qingre prescription, provided scientific basis for clinical medication.Methods:Collected hormone sensitive cases of children with nephrotic synrome from henan college of traditional medicine the frist affiliated hospital and children’s hospital of zhengzhou,and observed the 24h-UTP, ALB,TG,CHOL,IgG,FIB,PT,the side effects of hormore,routine blood,live and kidney fuction of safety indicators in the before and after treatment for 2weeks,4weeks,8weeks,12weeks,24weeks and 36 weeks.The last,compared the differece of two groups by tne statistical analysis.Results:①The treatment clinical curative effect of treatment group is better than the control group in the second week(P<0.05);②Treatment group recurrence rate was 9.4%,control group recurrence rate was 32.4%,the former is obviously lower than the latter(P<0.05);③Compared with the control group, the treatment group was better in reducing 24-UTP and increasing ALB,IgG from 2 weeks(P<0.05);④Compared with control group, treatment group was better in reducing CHOL and extending PT from 4 weeks(P<0.05);⑤TG and FIB wasn’t signficant statistical between the treatment group and the control group(P>0.05);⑥the treatment group has advantages in reducing infection frequency,sweating and impatience (P<0.05),but it wasn’t signcant differences in the full moon face and buffalo hump of the hormones’side efffects(P>0.05).Conclusion:①Yiqi Huayu Qingre prescription had the effects which were reducing urinary protein and increasing the role of plasma albumin and IgG,and it worked within 2weeks;②Yiqi Huayu Qingre prescription had the effects which were reducing cholesterol and FIB,and increasing PT,and it worked within 4weeks;③Yiqi Huayu Qingre prescription had the effects which were improving the body’s immune function,reducing the recurrence rate,infection frequency and reducing the side effects of hormones which were sweating and impatience,but it hadn’t effects which were reducing buffalo hump and moon face by hormones.Objective:Trough stimulused and induced in vitro podocyte damage model in mice by PAN,observed the effects of YIqi Huayu Qingre prescritition and its demolition party on mouse podocyte express podocin,and discuss Yiqi Huayu Qingre prescritition on podoocyte damage mechanism.Methods:Cultured cells in vitro and used RT-PCR to detect the expression of podocytes’podocin mRNA in different days and different concentration of PAN,and established the best concentration of PAN and the time of culturing cells;Afer establishing PAN model,podocytes were randomly divided into Yiqi group,Huayu group,Qingre group,Compound group,TW group,control group,model group and normal group,and they were cultured by using the medicated serum;The podocin mRNA and protein expressions of each group were detected by using RT-PCRand Wersten blot technology.Results:①The expression of podocin mRNA were higher than others on the seventh day and the eighth day(P<0.05),and they weren’t signficant statistical difference between the seventh day and the eighth day(P>0.05); ②Afer the PAN stimulused 24h,the expression of podocin mRNA were no significant statistical difference between the 50ug/ml group and 75ug/ml g Roup(P>0.05;)The expression of podocin mRNA of the rest groups were reduced with increasing congcentretion of PAN in podocyte damage 24h and 48h;③Compared with control group,the expresstion of podocin mRNA of compound group and TW group were higher in PAN intervention after 24h(P<0.05),but they were no difference(P>0.05);And compared with control group,Yiqi group,Huayu group and Qingre group were no diffference(P>0.05);④Comp-ared with control group,the expresstion of podocin mRNA of Huayu group,Qingre group,compound group and TW group were higher in PAN intervetion after 48h(P<0.05),compound group and TW group were better than Qingre group and Huayu group(P<0.05),but they were the same(P>0.05 );And compared with control group,Yiqi group was no difference(P>0.05); ⑤Compared control group,the expresstion of podocin protein of Yiqi group,Qingre group and compound group were higher(P<0.05),but Huayu group was no significant statistical difference(P>0.05) in PAN intervention after 24 hours;And the effect of compound group was better than Qingre group and Yiqi group(P<0.05),Yiqi group and Qingre group were the same (P>0.05);⑥Compared with control group,the expresstion of podocin protein of compound group and Huayu group were higher(P<0.05),but Yiqi group and Qingre group were no significant statistical difference(P>0.05)in PAN intervention after 48 hours;And the effect of compound group and Huayu group were the same(P>0.05).Conclusion:①This experriment conditions of model set up in the 50ug/ml PAN stimulation,and extracted the total RNA of cells in the seventh day;②After PAN stimulated podocyte for 24 hours,compound group could raise the expression of podocin mRNA and protein,Yiqi group and Qingre group could the expression of podocin protein,but they couldn’t raise the expression of podocin mRNA,And Huayu group couldn’t raise the expression of podocin mRNA and protein;③After PAN stimulated podocyte for 48 hours,compound group and Huayu group could raise the expression of podocin mRNA and protein,Qingre group could the expression of podocin mRNA,but they couldn’t raise the expression of podocin protein,And Yiqi group couldn’t raise the expression of podocin mRNA and protein.In addition,the effect of compound group was better than Huayu group and Qingre group. |
