Objective By lipopolysaccharide (LPS) lipopolysaccharide, stimulation, inductionof in vitro cultured rat glomerular mesangial cells (primary mesangial cells, MsC)proliferation, observing yiqi group of traditional Chinese medicine (composed ofradix astragali and dangshen), the blood group of traditional Chinese medicine(composed of salvia miltiorrhiza and leeches), heat group (composed of radixscutellariae and dandelion), the compound Chinese medicine set of traditionalChinese medicine (composed by all Chinese medicine) and tripterygium glycosidesexpression of rat MsC type p38mapk、Smad3and TGF-β1receptor comparative studyof effects of replenishing qi and blood circulation, clear heat medicine functionmechanism of proliferative glomerulonephritis.Methods In vitro culture of rat MsC, the use of lipopolysaccharide induce thehyperplasia, form the MsC proliferation model, will be made after the success of themould of MsC were randomly divided into normal group, model group and controlgroup, yiqi group and blood group,heat group,compound group, tripterygiumwilfordii group, respectively, to join all kinds of medicated serum continues todevelop, by RT-PCR method to detect each group containing medicine serum of MsCexpression p38MAPK mRNA influence;Using Wersten Blot method to detect eachgroup containing medicine serum of rat MsC expression p38MAPK,Samd3,TGF-β1effects.Results1.RT-PCR results showed: p38MAPK mRNA in the control group hadsignificant difference compared with model group(P <0.05);Groups of medicatedserum can inhibit the in vitro cultured MsC expression p38MAPK and Samd3,strongest compound group, compared with other medicated serum groups hadsignificant difference (P <0.05);Heat and blood group was no significant difference (P>0.05), but compared with yiqi group have significant differences(P<0.05);themedicated serum group compared with control group had significant difference (P<0.05).2.Wersten Blot results showed:p38MAPK、Samd3and TGF-β1protein in controlgroup and model group compared with significant difference (P <0.05);Groups ofmedicated serum can inhibit the in vitro cultured MsC expression p38MAPK、Samd3and TGF-β1, strongest compound group, compared with other medicated serumgroups had significant difference (P <0.05),Heat and blood group was no significantdifference (P>0.05), but compared with yiqi group have significant differences (P <0.05); The medicated serum group compared with control group had significantdifference (P <0.05).Conclusion Yiqi huayu qingre prescription and its party after the traditionalChinese medicinecan inhibit proliferation of MsC, reduce p38MAPK mRNAexpression, reduce p38MAPK、 Samd3and TGF-β1protein expression.Thusreplenishing qi, heat-clearing and Chinese medicine, Chinese traditional medicinecompound preparation of TCM for traditional Chinese medicine, blood circulation,tripterygium glycosides is a type of proliferative glomerulonephritis effectively drugs;Group in different groups of Chinese medicine, in the comparison of the compoundChinese medicine liquid, blood group and heat group Chinese medicine yiqi group isobviously better than the effect of traditional Chinese medicine, therefore, in thetreatment of proliferative glomerulonephritis should emphasis on promoting bloodcirculation to remove blood stasis, heat treatment of Yin.
