Objective: According to the quality standards in Chinese Pharmacopoeia (2010, Volume I),to study the technical standard of stir-baked with soil. Use the main contents as the indexes,to optimize the best processing technics of Radix Paeoniae Alba, Atractylodes, coix seed,Chinese yam, angelica stir-baked with soil, and determine the optimal processing parameters;make preliminary study on soil. Then make technical specifications and SOP for theprocessing method of stir-baked with soil. Provides the scientific basis for the drugsupervision and administration department manage Chinese medicine herbs; providesstandards for the development of Chinese medicine Pieces enterprises, and the qualityassurance for clinical use of pieces.Methods: Determine the contents of paeoniflorin and albiflorin by HPLC at once, make thecontents of paeoniflorin and albiflorin as indexes, and use central composite design/responsesurface methodology to optimize the best factors, such as temperature, time and the ratio ofradix paeoniae albe to terra flava usta. Determine the contents of Atractylenolide Ⅰ,Ⅲ byHPLC at once, to optimize the best processing technics. For Coix seeds, use orthogonalexperimental design and determine the content of three oleic acid glyceride by HPLC andELSD, to optimize the best processing technics. On the basis of the optimal processingtechnology of white peony root, Atractylodes, coix seed, make appearance, extract contentsas the indexes, the processing technologies of Chinese yam, and angelica are optimized. Theterra flava usta, halloysite, buff clay, sandy soil were fried and calcined, then determinate thepH with pH meter, blue absorption power of different samples. Extract10μm particles indifferent soil using the Stokes’s theorem, then use XRD to determinate mineral species in10μm particles.Results: For Radix Paeoniae Alba, response surface regression model fitted well, Theoptimal parameters are as follows: temperature200℃, time11min, the ratio of radixpaeoniae albe to terra flava usta is100:30. after Atractylodes fried with soil, atractylenolideⅠ, Ⅲ contents don’t immediately increase, but after comminuted and placed a month or soit’s contents reached the maximum. When Atractylodes is processed under heating in a pot at260℃for10min, it’s the best. For coix seed, the best processing method is: temperature 240℃, fried10min, pieces and stoves subsoil ratio is100:30. For Chinese yam, the betterprocessing method is: temperature260℃, fried10min, pieces and stoves subsoil ratio is100:25. For angelica, the better processing method is: temperature230℃, fried10min,pieces and stoves subsoil ratio is100:50. In different types of soil, stoves subsoil is acidity,other types of soils are alkaline, after fried the pH value decrease, after calcined but stovessubsoil, pH increased. The blue absorption power is no big changes after Halloysite fried orcalcined. For other types of soil, the crude and the fried adsorb methylene blue hardly, butafter calcined, blue absorption power has increased significantly, especially No.1buff clay.10μm particles content is the highest in No.4, followed by No.1, No.3, No.5, No.2.10μmparticles contents do not have too big difference between the crude and the fried, butcontents in calcined products significantly reduced. The result of XRD phase analysis are asfollows: No.1crude sample is mainly composed of quartz, illite, Albite,chlorite, kaolinite;No.2crude sample mainly contains: quartz, albite, chlorite, illite;NO.3crude sample mainlycontains: quartz, albite, chlorite, illite; NO.4crude sample mainly contains: quartz, hematite,muscovite, wustite, illite; NO.5crude sample mainly contains: quartz, albite, illite. Mineralspecies have no difference between the fried product and the raw. No.1calcined containsquartz, hematite, phengite, illite, montmorillonite; No.2calcined contains quartz, hematite,illite, albite (high temperature), Yakhontovite; No.3calcined is mainly composed of quartz,hematite, illite, Phengite, Yakhontovite; No.4calcined is mainly composed of quartz,hematite, illite, muscovite; No.5calcined contains quartz, illite, albite, halloysite.Conclusion: Optimized processing technics of Radix Paeoniae Alba is stable and reliable.The contents of paeoniflorin and albiflorin in processed products increased than rawproducts. After Atractylodes fried with soil is comminuted and sealed up, atractylenolide Ⅰ,Ⅲ contents will increase gradually with time. Processing temperature and time areimportant factors for the increase of atractylenolide Ⅰ, Ⅲ. The optimal processingtechnology of Coix seed is stable and reliable, three oleic acid glyceride increases afterprocessed. After different types of soil are calcined, the pH value increases and blueabsorption power increases significantly, and the species of clay mineral change. It may bethe reason why to use the stoves subsoil when pieces of Chinese Medicine stir-baked withsoil and why stir-baked with soil can invigorate the spleen to arrest diarrhea. Formulate theSOPs of Radix Paeoniae Alba, Atractylodes, coix seed, Chinese yam and angelica stir-bakedwith soil. The preliminary technical specification of stir-baked with soil has been formulated.
