| Object:The incidence of nonalcoholic fatty liver disease (NAFLD) is rising worldwide.It has been ranked as the second fastest growing disease in China. The relationshipbetween genetic susceptibility and the pathogenesis of NAFLD has been paid muchmore attention in recent years. The study of genes related to investigate itspathogenesis and treatment is very important. At present, multiple studies aboutpatatin like phospholipase domain containing3(PNPLA3) gene variation and thegenetic susceptibility to NAFLD has been confirmed abroad. But The correlationbetween polymorphism of PNPLA3gene and the incidence of NAFLD is still lessstudied in China, according to the differences of race and region, we analyzed theHan Ethnic Group’s polymorphism of PNPLA3in the Central Plains area, to explorethe PNPLA3genotype and allele frequency distribution, and the relationshipbetween PNPLA3gene rs738409and hereditariness to nonalcoholic fatty liverdisease. Method:Case control study, polymerase chain reaction (PCR) and gene sequencingmethod were used in the genotype polymorphism study of PNPLA3.85liver biopsydiagnosed cases of NAFLD patients and102cases healthy controls were detected inthe genotype polymorphism study of PNPLA3gene. The general features,biochemical results, liver stiffness values and histological results were collected inthe study. All the data were analyzed by SPSS17.0statistical package. TheHardy-Weinbeurg (H-W) equilibrium was used in the test of representativeness ofthe two groups; and the x2test, one-way analysis of variance and non-conditionalLogjstic regression were used to process experimental data.Result:In NAFLD and control groups, the frequency of CC genotype, CG genotype,and GG genotype for PNPLA3gene rs738409was16.47%,41.18%,42.35%and44.12%,41.18%,14.70%respectively. The G allele frequency was62.94%and35.29%for NAFLD and control groups respectively. There was significantdifference between the two groups (p<0.05). The Logistic regression analysissuggested that the OR of NAFLD was3.11(95%CI2.04-4.76) for GG genotypecompared with CC genotype. The level of BMI, TG, ALT, AST, FINS and liverstiffness values for GG genotype were higher than that of CC genotype, level offibrosis of fatty liver tissue GG genotype was higher than that of GC, CC genotype,GC genotype was higher than CC genotype, the difference was statisticallysignificant (p<0.05), while the differences of the degree of hepatic steatosis andinflammation compared between three genotypes were no statistically significant.Conclusion:The results showed that, the PNPLA3gene rs738409polymorphism withgenetic susceptibility to NAFLD is one of the important factors affecting theindividual NAFLD disease in the Han Ethnic Group in Central Plains area, and theallele of PNPLA3gene rs738409G is associated with the degree of fibrosis. |
