| Objective:To observe The effect of Abnormal Savda Munziq (ASMq) on cAMP-PKA and ERK signal pathway in the hippocampus of depression Rat models Methods:The84SD rats were randomly divided into6groups with14in each group:normal control group, depression animal model group, high, moderate and low ASMq granules dose groups (6.0,3.0,1.5g/kg) and the positive control group (Fluoxetine,3.5mg/kg). Except the normal control group, the rats in all other groups were given chronic unpredictable mild stress (CUMS) to build the depression animal models.The positive control group and the ASMq groups were orally administrated once a day for24days. The behavioral status of rats was evluated by Open-field test and sugar consumption experiment.24h after the last administration, all the rats were executed, the hippocampal tissue separated and cryopreserved for standby. The content of cAMP were tested with the method of ELISA. The protein expression of PKA, p-CREB, p-ERKl/2, ERK1/2in the hippocampus of rats were tested by Western-blot. Results:After CUMS, the body weight gain, sugar consumption, horizontal crossing numbers and erection times of depression group were significantly decreased than normal control group(P<0.01), so the depression model group was in an obvious depressive state. The body weight gain, sugar consumption, horizontal crossing numbers and erection times of positive control group, groups of ASMq granules for high(6.0g/kg), medium(3.0g/kg) were significantly increased than depression group (P<0.01).The content of cAMP, PKA and p-CREB protein expression levels in hippocampus of depression group were significantly lower than normal group(P<0.01, p<0.05). The content of cAMP, PKA and p-CREB protein expression levels in hippocampus of positive control group, moderate and high ASMq granules dose groups were significantly higher than depression group (P<0.01,P<0.05).The level of ERK1/2in each group were unchanged. The content of p-ERK1/2protein expression levels in hippocampus of depression group were significantly lower than normal group(P<0.01,P<0.05). The content of p-ERK1/2in positive control group, moderate(3.0g/kg) were significantly higher than depression group (P<0.01,P<0.05); Conclusion The anti-depression effect of Abnormal Savda Munziq (ASMq) may be related to regulating the cAMP-PKA and ERK signal pathway... |
