Tho Lnfluenoe Of Yishenjiangzhuo Prescr Lption On Chronio Renal Failure Rat Renal Tissue TGF-β1.BMP-7and Rat Kidney Cells

Objective:By observing YiShenJiangZhuo Prescription on the expression of5/6nephrectomy due to chronic renal failure rats renal tissue TGF-β1,BMP-7and proliferation,apoptosis of SD rat glomerular mesangial cells and renal interstitial fibroblasts culture in vitro to investigate the curative effect mechanism of YiShenJiangZhuo Prescription on chronic renal failure.Method:Adopt the methods of building animal model and cell culture.60clean7week-old male SD rats were divided into5groups including blank group,sham operation group,CRF model group,CRF control group(Niaoduqing granule group),CRF treatment group(Yishenjiangzhuo Prescription group).All groups built models by different ways.After4weeks,begin to give different medications and last for8weeks.Then observing the index TGF-β1.BMP-7in renal tissue by Immunohistochemistry.Observed objects were rat glomerular mesangial cells and renal interstitial fibroblasts culture in vitro.Preparation of normal rats drug-containing serum and CRF rats drug-containing serum.By cell medication experiment.the proliferation effect was evaluated bv MTT method.Result:1In CRF model group BUN and SCr increased significantly(P<0.05),TGF-β1elevated and BMP-7reduced in residual kidney(P<0.05).Observing renal tissue HE stain,glomerular volume increase,renal tubular atrophy,edema of renal tubular epithelial cells,renal interstitial hyperplasia of fibrosis,inflammatory cells infiltration.2Yishenjiangzhuo Prescription and Niaoduqing granule can reduce BUN and SCr in CRF rat serum(P<0.05),reduced TGF-β1and elevated BMP-7in residual kidney(P<0.05).Treatment of Yishenjiangzhuo Prescription is better than Niaoduqing granule.Observing renal tissue HE stain,they all can alleviate renal tissue fibrosis.3Yishenjiangzhuo Prescription drug-containing serum can inhibit the proliferation of SD rats glomerular mesangial cells and renal interstitial fibroblasts in vitro(P<0.05).Conclusion:1Yishenjiangzhuo Prescription can reduce TGF-β1expression and rise BMP-7 expression in chronic renal failure rats remnant renal tissue to improve the residual kidney pathological morphology and slow down the renal tissue fibrosis.2Yishenjiangzhuo Prescription drug-containing serum can inhibit the proliferation of glomerular mesangial cells and renal interstitial fibroblasts.3Yishenjiangzhuo Prescription can slow down the pathological development of chronic renal failure renal tissue fibrosis effectively.Yishenjiangzhuo Prescription is an effective medication in chronic renal failure prevention and cure.
