Aim:To explore the treatment principle of Yu Ping Feng San; observe the effect of Yu Ping Feng San extracted by different methods and to screen the best extraction method; investigate the anti-inflammatory mechanism of Yu Ping Feng San.Method:The data of Yu Ping Feng San was downloaded from Chinese BioMedical literature database. Human mononuclear macrophage U937 cells were induced to macrophages by PMA. The reasonable concentration of Yu Ping Feng San for the following experiments was studied by CCK-8 assay. After establishing inflammatory cell model, Yu Ping Feng San which extracted by ASE-DCM,ASE-MeOH, Decoction DCM, Decoction EtOAc, Decoction Butanol,Decoction Emulsion and Decoction Water respectively were added for 48h. The concentrations of TNF-α、IFN-γ and IL-1β from LPS-stimulated U937 cells were detected by ELISA.The acitvation of NF-κB and phosphorylation of JNK were detected by western blot.Results:Yu Ping Feng San was often used to treat diseases such as respiratory infections, asthma and cough. Traditional Chinese medicine (TCM) syndrome that Yu Ping Feng San treated was mainly correlated with deficiency and cold. Yu Ping Feng San was usually used together with Shen Qi pills, Si Jun Zi decoction and Sheng Mai San. For western medicines, Yu Ping Feng San was usually used together with cetirizine, interferon and antibiotics;The CCK-8 assay showed that different Yu Ping Feng San extracts didn’t exhibit cytotoxicity at the concentrations of 25μg/ml.. Subsequently, the concentration of 25p,g/ml was used for the further experiments. We found that the concentrations of TNF-α、IFN-γ、IL-1β released from LPS stimulated U937 cells increased significantly than those in the controls(P<0.01). After treatment with different extractions, the levels of TNF-α、IFN-γ、IL-1β in Decoction EtOAc group were significantly decreased when compared with other groups(P<0.01, P< 0.05).Western blot result showed that Yu Ping Feng San extracted by Decoction EtOAc remarkably suppressed NF-κB activation and down-regulated the phosphorylation of JNK in LPS-stimulated THP-1 cells.Conclusion:Yu Ping Feng San extracted by Decoction EtOAc showed the best anti-inflammatory activity, which was partly through regulating JNK/NF-κB signal pathways.
