ObjectiveTo Clear of hip fracture in elderly patients after acute lung injury does occur, inflammatory factors in the blood (IL-1β, IL-6, IL-10, TNF-α), the expression change of mtDNA.MethodsSelect 24 male aged SD rats as the experimental object, randomly divided into senile hip fracture and lung injury group (20 rats), blank control group (4 rats), after treatment respectively 4h,8h,24h,48h,72h.quantitative detection for inflammatory factor IL-1β, IL-6, IL-10, TNF-α, mtDNA test, were sacrificed after the animal, take the lung tissue, measurement of pulmonary permeability coefficient, water channel protein concentration, wet/ dry weight ratio of lung tissue and HE staining, to explore the relationship between mtDNA and inflammatory factors, and provides theoretical basis for the treatment of elderly hip fracture complicated with lung injury.Results1. hip fracture in the elderly rat peripheral blood IL-1 beta, IL-6, TNF- alpha levels increased with time gradually increased, reached the highest value to 24h, then decreased gradually, at each time point were significantly higher than that of the control group, IL-10 is the first drop to 24h to achieve the minimum value, and then gradually increased. Peripheral MR mtDNA in hip fracture trauma occurred after rising, and at 24h reached the peak, and then decreased, through correlation analysis found that, mtDNA and inflammatory factor IL-1 beta, IL-6 alpha, TNF- was positively correlated, negatively correlated with inflammatory factor IL-10.2. To compared to the control group, the aged hip fracture group lung tissue wet/dry weight ratio, lung permeability index increased significantly, and the difference has statistical significance (P<0.05), lung for Western blotting results showed that AQP-1 was significantly lower than the control group (P<0.05), lung tissue frozen section with HE staining lung tissue were observed under light microscope part tissue swelling, cytoplasm stained, increased cell size, alveolar septal thickening, more red blood cells and massive infiltration of neutrophils in the alveolar cavities lung pathological injury is, somewhat higher than the normal control group mice.Conclusions1. Hip fracture in aged rat peripheral blood IL-1, IL-6, TNF- alpha beta content with the passage of time gradually increased and reached the peak on 24h, and in the subsequent trend of gradual decline, the content of IL-10 with time is first reduced and increased gradually to the lowest value after 24h.2. Aged rats suffered after a hip fracture can cause acute lung injury, lung tissue wet weight ratio increased, increased lung permeability coefficient, the expression level of AQP-1 decreased, pathological manifestations of pulmonary edema, alveolar hemorrhage, inflammatory cell infiltration, lung organization occur disorder.3. mtDNA increased in the hip fracture in aged rats, there was a positive correlation between the occurrence and inflammatory reaction.
