| PurposeTo observe the expression of SPARC, E-cadherin and EZH2in proliferative phase endometrium (PPE), simple endometrial hyperplasia.(SEH), complex endometrial hyperplasia (CEH), atypical endometrial hyperplasia (AEH) and endometrioid adenocarcinoma (EA) specimens, investigate the relationship between the three proteins and clinicopathological characters of EA and the EA occurrence and development role.MethodsThe expression of SPARC, E-cadherin and EZH2was detected in PPE,SEH,CEH.AEH and EA tissues by immunohistochemical staining (streptavidine-peroxidase method).Results1. The positive expression rate of SPARC and E-cadherin in EA tissues was29.23%and27.69%, respectively, significantly lower than that in PPE,SEH.CEH.AEH (P<0.01);While the positive expression rate of EZH2in EA tissues was73.85%, respectively, significantly higher than that in PPE,SEH,CEH,AEH tissues (P<0.0l).2. The expression of SPARC and E-cadherin was associated with histopathology grading, myometrial infiltration and lymph node metastasis in EA (P<0.05).Expression of EZH2was associated with histopathology grading, myometrial infiltration (O<0.05), but not associated with lymph node metastasis in EA (P>0.05).3. The expression of SPARC and E-cadherin was positive related (rs=0.226, P<0.05). Expression of SPARC and EZH2was negatively related(rs=-0.201, P<0.05).Expression of E-cadherin and EZH2were negatively related in EA (rs=-0.214, P<0.01).Conclusion1. Expression of SPARC and E-cadherin at the protein level is significantly decreased from PPE,SEH,CEH,AEH to EA,while expression of EZH2is significantly increased. This suggests that their expression may associate with occurrence of EA. 2. Expression of SPARC,E-cadherin and EZH2is closely associated with histopathology grading,myometrial infiltration in EA.Expression of SPARC and E-cadherin is closely associated with lymph node metastasis. This suggests that their different expression may play a key role in development, infiltration and metastasis of EA.3. Expression of SPARC, E-cadherin and EZH2in EA indicates that their expression may exist within a kind of interaction. Combined detection of SPARC, E-cadherin and EZH2will contribute to the clinical diagnosis,treatment and efficacy evaluation of EA. |
